    Isolamento e identificação da bactéria anaeróbia Clostridium perfringens no veneno e quelíceras de Loxosceles intermedia (aranha marrom) : avaliação da potencialização da lesão dermonecrótica decorrente dos acidentes loxoscélicos

    Abstract
    Resumo: Loxoscelismo, ou envenenamento pela aranha marrom (Loxosceles intermedia), pode resultar, no local da picada, extensa dermonecrose, hemorragia e, eventualmente, reações sistêmicas que podem ser letais. A partir de aranhas coletadas na natureza, utilizando-se do aparelho venenífero (quelíceras e glândulas produtoras de veneno), e de culturas microbiológicas realizadas em condições aeróbias, facultativas e anaeróbias, foram feitos isolamento e identificação de diversas bactérias, dentre elas a bactéria anaeróbia Clostridium perfringens. Esta bactéria é de grande interesse médico em função de seu envolvimento em patologias dermonecrotizantes e sistêmicas. Utilizando-se o veneno de Loxosceles intermedia (espécie que prevalece na região de Curitiba), associado com Clostridium perfringens, em modelos experimentais de dermonecrose em coelhos, foi possível identificar que tal conjugação, veneno e bactéria resultou em aumento exacerbado do quadro dermonecrótico comparado àquele induzido somente pela inoculação do veneno. Essa resposta sugere a participação de Clostridium perfringens no quadro dermonecrótico grave dos acidentados e abre possibilidades para a adição profilática da antibioticoterapia como abordagem terapêutica adicional no tratamento do loxoscelismo.
     
    Abstract: Loxoscelism or the envenoming by the brown spiders (Loxosceles genus spiders), may produce extensive dermonecrosis and hemorrhage at the bite site and, eventually, systemic reactions that may be lethal. Isolation and identification of many different bacteria, among them Clostridium perfringens, of great medical importance due to its involvement in dermonecrotizing and systemic alterations, was carried out through microbiological cultures in aerobic, facultative and anaerobic conditions, from the venomous apparatus (fangs and venom producing glands) of Loxosceles intermedia spiders obtained directly from nature. In the present work we investigate the time course of dermonecrotic lesion induced in rabbits by inaculation of venom alone and conjugated with Clostridium perfringens. The results obtained from experiments indicated that spider venom and anaerobic bacteria conjugation was able to cause a striking increase of the dermonecrotic picture when compared to venom alone. It suggest a important role for Clostridium perfringens in the dermonecrotic lesion evolution in accidented patients. Also, it opens the possibility for the adapting of antibiotic therapy in treating loxoscelism.
