    Padronização de uma técnica de PCR-SSP para identificação de genótipos do gene da BCHE

    Abstract
    Resumo : As colinesterases, acetilcolinesterase (AChE) e butirilcolinesterase (BChE) são enzimas que realizam a hidrólise de ésteres de colina, sendo o principal substrato para tais enzimas o neurotransmissor acetilcolina (ACh). A BChE realiza igualmente a hidrólise de outros ésteres como a butirilcolina (BuCh). Alguns indivíduos podem apresentar mutações em porções específicas do gene da BCHE, manifestando uma alta sensibilidade quando expostos a determinados fármacos, como a succinilcolina, fazendo com que baixas administrações de tal medicamento sejam suficientes para entrada em estado de apnéia por algumas horas. Tendo em vista a importância farmacogenética da identificação prévia de indivíduos com variações na expressão da BChE, o presente estudo desenvolveu a padronização da técnica de PCR-SSP, por meio do desenho de quatro primers para cada um dos genótipos mutantes A, K e 116. Foi possível padronizar efetivamente a técnica para as mutações A e -116, sendo necessários maiores estudos acerca da aplicação da técnica para a mutação K
     
    Abstract : The cholinesterases, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are enzymes responsible for the hydrolysis of choline esters. Their main substrate is the neurotransmitter acetylcholine (ACh). BChE also performs the hydrolysis of other esters such as butyrylcholine (BuCh). Some individuals may have mutations in specific portions of the BCHE gene, manifesting a high sensitivity when exposed to certain drugs, such as succinylcholine, low administrations of this drug in thoses cases are sufficient to enter a state of apnea for a few hours. Considering the pharmacogenetic importance of the prior identification of individuals with variations in BChE expression, the present study developed the standardization of the PCR-SSP technique, through the design of four primers for each of the mutant genotypes A, K and -116. It was possible to effectively standardize the technique for the A and -116 mutations, requiring further studies on the application of the technique for the K mutation
