Avaliação do propólis de abelhas Tetragona clavipes na sensibilização de células MDA-MB-231 à citotoxicidade induzida por quimioterápicos

Resumo: O câncer de mama é a neoplasia mais incidente em mulheres, bem como a responsável pela maior mortalidade associada ao câncer no sexo feminino. Dentre os vários tipos moleculares de câncer de mama, destacam-se tumores triplo negativos, com o pior prognóstico e a menor sobrevida global, visto que pacientes portadores desse tipo de tumor frequentemente apresentam metástases, quimiorresistência e recidiva da doença. Isso evidencia a necessidade de se buscar novas abordagens terapêuticas, mais efetivas e seletivas para o tratamento dessa doença. Nesse sentido, merece destaque o própolis, o qual vem sendo amplamente investigado devido ao seu potencial citotóxico e antitumoral. Este trabalho visa avaliar o própolis da abelha Tetragona clavipes (BORÁ) como agente capaz de sensibilizar células de carcinoma mamário à ação de doxorrubicina (DOXO), paclitaxel (PACLI) e 5-fluorouracil (5-FU). A seleção da linhagem tumoral MDA-MB-231 para a execução deste trabalho baseou-se em resultados obtidos a partir do screening realizado por meio do ensaio do MTT, que demonstrou que BORÁ apresentou citotoxicidade significativa em um painel de linhagens tumorais, sobretudo na linhagem de câncer de mama triplo negativa. Ainda, os resultados obtidos evidenciaram que tal citotoxicidade foi seletiva, comparando-se os efeitos induzidos por BORÁ nas células MDA-MB-231 e na linhagem não tumoral L929. Em relação a avaliação da capacidade de BORÁ sensibilizar seletivamente células MDA-MB-231, a ação desses três quimioterápicos, resultados promissores foram obtidos com as associações 5-FU (0,125 - 4 µM) + BORÁ (22,51 µg/mL), sendo o esquema de pré-tratamento, o mais efetivo. O ensaio de incorporação de iodeto de propídeo/laranja de acridina mostrou que a associação 5-FU (1 µM) + BORÁ (22,51 µg/mL), aplicada na forma de pré-tratamento, induziu morte celular por mecanismos que atuam na ruptura/desorganização da membrana celular, enquanto o ensaio do wound healing mostrou que tal combinação reduziu a migração de células MDA-MB-231, sugerindo que a mesma apresenta potencial antimetastático. Já o ensaio clonogênico demonstrou que as associações 5-FU (1 µM) + BORÁ (22,51 µg/mL) e 5-FU (2 µM) + BORÁ (22,51 µg/mL) induziram efeito anticlonogênico seletivo, estando tal efeito associado ao potencial antiproliferativo das combinações, bem como ao fato de reduzirem a capacidade de adesão celular. Por fim, o ensaio do CPD (cumulative population doubling) mostrou que a longo prazo, o tratamento mais promissor foi a associação 5-FU (1 µM) + BORÁ (22,51 µg/mL), o qual deve ser estendido até o 24º dia, intervalo de tempo no qual a associação retardou de forma seletiva e significativa, o crescimento celular, ao evitar a recuperação total da taxa de proliferação de células MDA-MB-231.Dessa forma, os resultados obtidos sugerem que o pré-tratamento de células MDA-MB-231 com BORÁ potencializa a ação citotóxica induzida por 5-FU a curto e longo prazo. Abstract: Breast cancer is the most frequent neoplasm in women, and responsible for the highest cancer-associated mortality in females. Among the various molecular types of breast cancer, triple negative tumors stand out, which have a worse prognosis and lower overall survival, since patients with this type of tumor often have metastases, chemoresistance, and disease recurrence. This highlights the need to search for new, more effective, and selective therapeutic approaches for the treatment of this disease. In this way, propolis deserves to be highlighted, it has been widely investigated due to its cytotoxic and antitumor potential. Therefore, this work aims to evaluate whether Tetragona clavipes bee propolis (BORÁ) is capable of sensitizing mammary carcinoma cells to the action of doxorubicin (DOXO), paclitaxel (PACLI), and 5-fluorouracil (5-FU). The selection of the MDA-MB-231 tumor line for this work was based on the results obtained from a screening carried out using the MTT assay, which showed that BORÁ presented significant cytotoxicity in a panel of tumor lines, especially in the triple-negative breast cancer. Furthermore, the results obtained showed that its cytotoxicity was selective, comparing the effects induced by BORÁ in MDA-MB-231 cells and the non-tumor lineage L929. Regarding the evaluation of the ability of BORÁ to selectively sensitize MDA-MB-231 cells to the action of the three chemotherapy agents mentioned, promising results were obtained with the associations 5-FU (0.125 - 4 µM) + BORÁ (22.51 µg/mL), being the most effective pretreatment regimen. The propidium iodide/acridine orange incorporation assay showed that the association 5-FU (1 µM) + BORÁ (22.51 µg/mL), as a pretreatment induced cell death by mechanisms that act on rupture/ disorganization of the cell membrane. The wound healing assay showed that this combination reduced the migration of MDA-MB-231 cells, suggesting that it has antimetastatic potential. The clonogenic assay showed that the associations between 5-FU (1 µM) + BORÁ (22.51 µg/mL) and 5-FU (2 µM) + BORÁ (22.51 µg/mL) induced a selective anticlonogenic effect, being this effect associated with the potential antiproliferative effect of the combinations, as well as the fact that they reduce the cell adhesion capacity. Finally, the CPD (cumulative population doublings) trial showed that in the long term, the most promising treatment was the association 5-FU (1 µM) + BORÁ (22.51 µg/mL), which should only be extended until the 24th day, an interval of time in which the association selectively and significantly delayed cell growth, preventing full recovery of the proliferation rate of MDAMB- 231 cells. Thus, the results obtained suggest that MDA-MB-231 cells pretreated with BORÁ potentiate the cytotoxic action induced by 5-FU in short and long term.
