Estudio fitoquímico, actividades biológicas y antioxidantes de las hojas, tallo y del aceite esencial de la especie Allophylus semidentatus (Miq.) Radlk. (Sapindaceae)

Resumo: A espécie Allophylus semidentatus (Miq.) Raldk. pertence à família Sapindaceae, nativa do Brasil, mas não endêmica, tendo ocorrência em todas as regiões do país. A falta de estudos nesta espécie foi a principal razão para o desenvolvimento deste trabalho, o qual foi realizar o estudo fitoquímico, avaliar as propriedades antioxidantes e atividades biológicas. Estudos realizados em outras espécies do gênero Allophylus tiveram propriedades antiinflamatórias, antimaláricas, antiulcerogênicas, analgésicas entre outras. O material vegetal foi coletado na cidade de Curitiba no Brasil e identificado por um especialista do Museu Botânico Municipal de Curitiba. Na análise fitoquímica inicial, constatou-se a presença de alcaloides, compostos fenólicos e triterpenos. O óleo essencial foi extraído das folhas das espécies com uma equipe de Clevenger, obtendo-se um rendimento de 0,73% apresentando como principais compostos y- muuroleno, g-cadineno e Z-cariofileno. Os extratos brutos das folhas e caules foram obtidos utilizando etanol como solvente e utilizando um equipamento de Soxhlet. Posteriormente, este extrato bruto foi fracionado com solventes de polaridade crescente para obtenção das frações de hexano, clorofórmio, acetato de etila e remanescente, através de um equipamento Soxhlet modificado. A partir da fração hexanólica do caule, foi isolado e identificado a alantoína e friedelina. Atividades biológicas e antioxidantes foram realizadas em extratos brutos, frações e óleo essencial. No teste de toxicidade in vitro em Artemia salina, as frações hexanólica e clorofórmica do caule foram consideradas pouco tóxicas. Nos ensaios da atividade antioxidante pelo método de formação do complexo fosfomolibdeno, a fração acetato de etila do caule apresentou uma atividade de 151,55% em relação ao padrão de rutina, enquanto a mesma fração, estudada pelo método de redução do radical DPPH apresentou a maior atividade, com IC50 de 9,20yg/mL. No teste de compostos fenólicos totais, novamente a maior atividade foi observada na fração acetato de etila do caule (135,06±0,44mg EAG/g), enquanto o óleo essencial apresentou um valor de (30,17±0,29mg EAG/g). Na atividade alelopática, evidenciou-se a influência negativa de vários parâmetros medidos em Lactuca sativa, especialmente aqueles registrados pelo óleo essencial. Abstract: The Allophylus semidentatus (Miq.) Radlk. specie belong to the family of Sapindaceae, which is native to Brazil but not endemic. It is confirmed to have been found in all regions of the country. The lack of studies in this specie was the main reason to this work, which it is the aim to perform phytochemical studies. In this sense, we evaluate the antioxidant properties and biological activities. In previous studies, other species of the genus Allophylus had indicated the presence of anti-inflammatory, antimalarial, antiulcerogenic, analgesic properties and others properties. The vegetal material was collected in the Curitiba city (Brazil), and was identified by a specialist of the Municipal Botanical Museum of the same city. In the initial phytochemical analysis, the presence of alkaloids, phenolic compounds and triterpenes was evidenced. With a Clevenger's equipment, the essential oil was extracted from leaves of the species. From this type of essays, we obtaining a efficiency of 0.73%, showing as majority compounds: y-muurolene, g-cadinene and Z-caryophyllene. The raw extract was obtained using ethanol as solvent and using a Soxhlet equipment. Later, this extract was fractionated by using solvents of increasing polarity and a modified Soxhlet equipment, to obtain the fractions of hexane, chloroform, ethyl acetate and remanescent. From the hexanolic fraction of the stem, allantoin and friedelin were isolated and identified. Biological and antioxidant activities were performed on raw extract, fractions and essential oil. In the in-vitro toxicity test to the Artemia salina, hexanolic and chloroformic fractions of stems were found slightly toxic. In the tests of antioxidant activity, by means of the formation of the fosfomolibdeno complex method, the ethyl acetate fraction of stems presented an activity of 151.55%, compared with the routine pattern. Whereas that the same fraction studied by means of reduction of the DPPH radical method showed the highest activity, with an IC50 of 9.20 yg/mL. In the test of total phenolic compounds, the fraction of ethyl acetate of the stem (135.06±0.44mg EAG/g) was who presented a high value again; whereas that the essential oil presented a value of (30.17±0.29mg EAG/g). In the allelopathic activity, the negative influence on several of the parameters measured in Lactuca sativa was evidenced, particularly by the essential oil registers.
