Estudo fitoquímico, atividades biológicas, propriedades antioxidantes e antimicrobiana da espécie Myrcia venulosa DC, Myrtaceae

Resumo: A espécie Myrcia venulosa (Myrtaceae) é nativa e endêmica do Brasil, com ocorrência na Mata Atlântica. O objetivo deste trabalho foi realizar o estudo fitoquímico e avaliar as propriedades antioxidantes e atividades biológicas. O material vegetal foi coletado em Curitiba, Paraná, Brasil e identificado no setor de engenharia florestal da UFPR. As folhas secas foram utilizadas para o preparado do extrato hidroalcoólico e suas respectivas frações. Na triagem fitoquímica foi observada a presença de compostos fenólicos e esteroides e/ou triterpenos. O óleo essencial foi extraído por Clevenger, resultou em um rendimento de 0,22%, cujo compostos majoritários foram Germacreno-B (13,77%), óxido de cariofileno (16,23%) e B-cariofileno (6,12%). A extração das folhas e galhos foi realizada por Soxhlet e após o fracionamento, resultaram-se as frações hexano, clorofórmio, acetato de etila e remanescente. No teste antioxidante por formação do complexo fosfomolibdênio as amostras folha acetato de etila, folha remanescente e óleo essencial apresentaram atividadade superior ao BHT e rutina. No teste redução do radical DPPH a amostra com melhor resultado foi o óleo essencial (CL 50 5,88 Mig/mL). A determinação de fenóis totais e doseamento de flavonoides apontou significativo resultado para fração folha acetato de etila (51,15 mg.EAG/g) e óleo essencial (57,17 mg.EAG/g) , sendo essas as amostras que mais se destacaram. No ensaio de toxicidade in vitro frente à Artemia salina foi constatado toxicidade moderada do oleo essencial (CL50 372,28 Mig/mL) e baixa da fração folha acetato de etila (CL50 679,25 Mig/mL) e fração galho acetato de etila (CL50 953,09 Mig/mL). Obteve-se atividade hemolítica em relação ao óleo essencial de 57,46% na concentração de 1000 Mig/mL. Na avaliação da atividade antimicrobiana frente às bactérias Enterococcus faecalis, Streptococcus pyogenes, Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Proteus mirabilis e Enterobacter aerogenes ocorreu inibição do crescimento bacteriológico. Pela metodologia da microdiluição, os extratos também exibiram atividade antimicrobiana, mas com valores de CIM demonstrando uma atividade moderada a fraca. Abstract: The species Myrcia venulosa (Myrtaceae) is native and endemic to Brazil, occurring in the Atlantic Forest. The objective of this work was to perform the phytochemical study and to evaluate the antioxidant properties and biological activities. The plant material was collected in Curitiba, Paraná, Brazil and identified in the forest engineering sector of UFPR. The dry leaves were used for the preparation of the hydroalcoholic extract and their respective fractions. In phytochemical screening the presence of phenolic compounds and steroids and / or triterpenes were observed. The essential oil was extracted by Clevenger, resulting in a yield of 0.22%, whose major compounds were Germacrene-B (13.77%), caryophyllene oxide (16.23%) and B-caryophyllene (6.12%). Leaf and branch extraction was performed by Soxhlet and after fractionation the hexane, chloroform, ethyl acetate and remainder fractions resulted. In the antioxidant test due to the formation of the phosphomolybdenum complex the ethyl acetate leaf, remnant leaf and essential oil samples showed superior activity to BHT and routine. In the DPPH radical reduction test the best-performing sample was essential oil (LC 50 5.88 Mig / mL). The determination of total phenols and flavonoid assay showed a significant result for ethyl acetate leaf fraction (51.15 mg.EAG / g) and essential oil (57.17 mg.EAG / g), which were the most outstanding samples . In the in vitro toxicity test against Artemia salina, moderate toxicity of essential oil (LC50 372.28 Mig / mL) and low fraction of ethyl acetate sheet (LC50 679.25 Mig / mL) and fraction ethyl acetate LC50 953.09 Mig / mL). Hemolytic activity was obtained in relation to the essential oil of 57.46% in the concentration of 1000 Mig / mL. In the evaluation of the antimicrobial activity against the bacteria Enterococcus faecalis, Streptococcus pyogenes, Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Proteus mirabilis and Enterobacter aerogenes occurred inhibition of bacteriological growth. By microdilution methodology, the extracts also showed antimicrobial activity, but with MIC values demonstrating a moderate to weak activity.
