Etiologia e distribuição espacial de cancros basais em Eucalyptus benthamii no sul do Brasil

Resumo: O Eucalyptus benthamii é uma espécie promissora devido à maior tolerância às geadas na região Sul do Brasil em relação às demais espécies plantadas no país. No entanto, o aumento da área plantada com essa espécie levou ao aumento de relatos de doenças, destacando-se o cancro basal. Este trabalho teve como objetivo identificar e caracterizar os patógenos associados ao cancro basal ocorrente em duas áreas de testes de progênies de E. benthamii, em Mafra, SC e São Mateus do Sul, PR, através do isolamento, caracterização morfológica das colônias e caracterização molecular dos isolados com a filogenia das regiões ITS e ß-tubulina utilizando o método de Inferência Bayesiana. Foram realizados testes de patogenicidade em árvores e em brotações destacadas de E. benthamii. Também foram determinadas as temperaturas ideais para crescimento micelial em meio de cultura e de infecção em brotações destacadas e E. benthamii. A distribuição espacial foi analisada através da distribuição da incidência e severidade da doença em campo e com o cálculo de Índice de Dispersão. As espécies identificadas neste estudo através da filogenia foram Neofusicoccum dianense, Neofusicoccum parvum, Neofusicoccum spp., Pestalotiopsis telopeae, Pestalotiopsis trachycarpicola, Pestalotiopsis chamaeropis, Neopestalotiopsis formicarum, Neopestalotiopsis sp. e Cytospora eucalypticola. Todos os isolados foram patogênicos nos testes de patogenicidade. A melhor faixa de temperatura para o crescimento micelial dos isolados de Neofusicoccum, Pestalotiopsis e Neopestalotiopsis foi de 20 a 28 oC, enquanto para os isolados de C. eucalypticola foi de 24 a 32 oC. A melhor temperatura para infecção de isolados de Neofusicoccum, P. trachycarpicola e Neopestalotiopsis formicarum em brotações destacadas de E. benthamii foi 28 oC, enquanto para o isolado de Neopestalotiopsis sp foi 24 oC e para os isolados de C. eucalypticola foi 32 oC. A incidência do cancro basal na área de Mafra foi de 47,06% e na área de São Mateus do Sul foi de 47,76%. A severidade foi maior na área de São Mateus do Sul, e ambas as áreas apresentaram Índice de Dispersão do tipo agregado. Nos testes de patogenicidade, os isolados de Neofusicoccum apresentaram maior agressividade em relação aos de Cytospora, Pestalotiopsis e Neopestalotiopsis, e os isolados de Neofusicoccum apresentaram sintomas similares aos sintomas observados em campo. Abstract: Eucalyptus benthamii is a promising species due to its greater frost tolerance in southern Brazil, compared to other species planted in the country. However, the increase in planted area, with this species, has led to an increase in diseases reports, particularly basal canker. The objective of this work was to identify and characterize the pathogens associated with basal canker, occurring in two E. benthamii progeny test areas, in Mafra SC and São Mateus do Sul PR, through isolation, morphological characterization of colonies and molecular characterization of isolates with the phylogeny of the ITS and ß-tubulin regions using the Bayesian Inference method. Pathogenicity tests were performed on trees and on detached shoots of E. benthamii. The ideal temperatures for mycelial growth in culture media and infection in detached shoots and E. benthamii were also determined. The spatial distribution was analyzed by distributing disease incidence and severity in the field and calculating the dispersion index. The species identified in this study through phylogeny were Neofusicoccum dianense, Neofusicoccum parvum, Neofusicoccum sp., Pestalotiopsis telopeae, Pestalotiopsis trachycarpicola, Pestalotiopsis chamaeropis, Neopestalotiopsis formicarum, Neopestalotiopsis sp. and Cytospora eucalypticola. All isolates were considered pathogenic in pathogenicity tests. The best temperature range for mycelial growth of Neofusicoccum, Pestalotiopsis and Neopestalotiopsis isolates was 20 to 28 oC, while for C. eucalypticola isolates it was 24 to 32 oC. The best temperature for infection of isolates of Neofusicoccum, P. trachycarpicola and Neopestalotiopsis formicarum in detached shoots of E. benthamii was 28 oC, while for the isolate of Neopestalotiopsis sp it was 24 oC and for the isolates of C. eucalypticola it was 32 oC. The basal canker incidence in Mafra area was 47.06% and in São Mateus do Sul area it was 47.76%. The severity was greater in São Mateus do Sul area, and both areas presented an aggregate dispersion index. In pathogenicity tests, Neofusicoccum isolates showed greater aggressiveness compared to Cytospora, Pestalotiopsis and Neopestalotiopsis, and Neofusicoccum isolates showed symptoms similar to those observed in the field.
