Síntese de 2-(organocalcogenil)benzo[b]calcogenofenos via ciclização nucleofílica intramolecular promovida por ácidos de Bronsted-Lowry

Resumo: O presente trabalho consiste no desenvolvimento de uma metodologia para a obtenção de 2-(organocalcogenil)benzo[b]calcogenofenos em poucas etapas reacionais, utilizando condições ambientalmente amigáveis e calcogenoalquinos substituídos com calcogenoéteres (S, Se e Te) na posição orto através de ciclizações nucleofílicas intramoleculares promovidas por eletrófilos. A melhor condição reacional alcançada para a obtenção de 2-(organocalcogenil)-benzo[b]calcogenofenos utiliza o ácido p-toluenossulfônico (1,0 equivalente) em etanol (2,0 mL) sob aquecimento de 120 ºC durante 24 h, obtendo-se o 2-(fenilsulfanil)benzo[b]tiofeno em 82% de rendimento. Por meio dessa metodologia, foi possível preparar uma série de derivados de 2-(organocalcogenil)-benzocalcogenofenos com rendimentos de razoáveis a excelentes. Primeiramente foram sintetizados os 2-(organosulfanil)benzo[b]tiofenos com rendimentos entre 63 a 93% (7 exemplos), assim como 2-(organoselanil)benzo[b]tiofenos com rendimentos entre 57 a 94% (8 exemplos). Ainda foram preparados o 2-(fenilsulfanil)benzo[b]selenofeno e o seu correspondente contendo átomo de selênio no anel exocíclico, 2-(fenilselanil)benzo[b]selenofeno, com rendimentos de 97 e 93%, respectivamente, no total foram sintetizados 19 exemplos, sendo 4 inéditos. Buscando investigar aplicação sintética do 2-(fenilsulfanil)benzo[b]tiofeno (2a), foram realizadas reações de acoplamento cruzado e calcogenilações, que permitiram a síntese do 2-(fenilsulfanil)-3-(fenilselanil)benzo[b]tiofeno em 68% e do 2-(fenilsulfanil)-3-(fenil)benzo[b]tiofeno em 52%. Em relação ao desenvolvimento de uma metodologia one-pot partindo do 1-(2,2-dibromovinil)-2-(butiltio)benzeno (3a), observou-se a formação de dois produtos reacionais, o 2-(fenilsulfanil)benzo[b]tiofeno (2a) e 3-bromo-2-(fenilsulfanil)benzo[b]tiofeno (2aa), sendo obtido preferencialmente o segundo em relação ao primeiro. Posteriormente foi otimizado a formação de 2aa, aplicando na segunda etapa 5,0 equivalentes de ácido p-toluenossulfônico, aquecimento de 60 ºC durante 24h, dessa maneira se obteve o 3-bromo-2-(fenilsulfanil)benzo[b]tiofeno (2aa) em 92%. Abstract: The present work consists in the development of a synthetic methodology to obtain 2-(organochalcogenyl)benzo[b]chalcogenophenes in few reaction steps using environmentally friendly conditions and chalcogenalkynes substituted with chalcogenethers (S, Se and Te) in the ortho position through electrophilic-promoted intramolecular nucleophilic cyclizations. In view of the reactivity of alkynes in 5-endo-dig cyclizations, Bronsted-Lowry acids were used as electrophiles in the cyclization in an unprecedented way. The best reaction condition achieved to obtain 2-(organochalcogenyl)benzo[b]chalcogenphenes uses p-toluenesulfonic acid (1.0 equivalent) in ethanol (2.0 mL) under heating at 120 °C for 24 h, obtaining 2-(phenylsulfanyl)benzo[b]thiophene in 82% yield. Through this methodology, it was possible to prepare a series of derivatives of 2-(organochalcogenyl)-benzochalcogenphenes with reasonable to excellent yields. First, 2-(organosulfanyl)benzo[b]thiophenes were synthesized with yields between 63 and 93% (7 examples), as well as 2-(organoselanyl)benzo[b]thiophenes with yields between 57 and 94% (8 examples). 2-(phenylsulfanyl)benzo[b]selenophene and its correspondent containing selenium atom in the exocyclic ring, 2-(phenylselanyl)benzo[b]selenophene, were also prepared with yields of 97 and 93% respectively. Seeking to investigate the synthetic application of 2-(phenylsulfanyl)benzo[b]thiophene (2a), cross-coupling reactions and chalcogenylations were carried out, which allowed the synthesis of 2-(phenylsulfanyl)-3-(phenylselanyl)benzo[b]thiophene in 68% and of 2-(phenylsulfanyl)-3-(phenyl)benzo[b]thiophene in 52%. Regarding the development of a one-pot methodology starting from 1-(2,2-dibromovinyl)-2-(butylthio)benzene (3a), it was observed the formation of two reaction products, 2-(phenylsulfanyl)benzo[b]thiophene (2a) and 3-bromo-2-(phenylsulfanyl)benzo[b]thiophene (2aa), the latter being preferably obtained over the former. Subsequently, the formation of 2aa was optimized, applying 5.0 equivalents of p-toluenesulfonic acid in the second step, heating at 60 °C for 24 hours, thus obtaining 3-bromo-2-(phenylsulfanyl)benzo[b]thiophene (2aa) by 92%.
