Análise microestrutural de apatita de dentes humanos : estimativa de idade e sexo de indivíduos para apicações forenses

Resumo: Os dentes são os órgãos mais duradouros do corpo humano, sendo formados por minerais do grupo da apatita, principalmente Ca5(PO4)3OH, podendo haver em sua estrutura de 4% a 6% de ânions carbonato (CO3)²-. Uma das vertentes das pesquisas em Ciências Forenses baseia-se em refinar processos de identificação de cadáveres e neste contexto diversas técnicas têm sido aplicadas ao dente com o objetivo de se obter inferências sobre a idade de indivíduos. A obtenção do tamanho médio dos cristalitos de dentes pela difratometria de raios X (DRX) é comumente empregada, partindo-se da premissa de que há uma variação no tamanho dos cristalitos médio em função da idade. Outras técnicas também têm sido empregadas, embora ainda de maneira pouco difundida. Esta pesquisa foi realizada em trinta e sete dentes humanos, de doadores consentidos, onde os dentes foram concentrados em três grandes grupos: permanentes terceiros molares, permanentes não terceiros molares e decíduos. Os dentes foram analisados por difratometria de raios X, por microscopia eletrônica de varredura (MEV) e microssonda eletrônica. Nas análises por DRX os difratogramas obtidos para cada dente foram agrupados para uma comparação entre cada tipo de dente e para um mesmo tipo de dente em diferentes indivíduos; os resultados foram refinados pela equação de Scherrer para a obtenção do tamanho médio do cristal. Na microscopia eletrônica de varredura, os dentes foram analisados em elétrons secundários, onde foram observados aspectos morfológicos como os túbulos dentinários, quantificados por área de quadrícula e tamanho dos diâmetros médios. Em EDS foram mapeados os elementos Ca, P, Mg e C. Foram, também, analisados os óxidos P2O5, CaO, Na2O, K2O, MgO, SiO2, SO3, SrO, Al2O3 e os elementos F e Cl, em microssonda eletrônica. De acordo com os resultados, há uma relação negativa entre tamanho de cristalito e idade de indivíduos. Os dentes mais novos (decíduos) apresentam cristalitos médio em torno de 50 nm, enquanto os dentes de maior idade possuem tamanho médio do cristal entre 30 e 40 nm. Os dentes terceiros molares apresentaram um tamanho médio superior ao esperado para dentes permanentes (60nm). Na contagem de túbulos dentinários por área de quadrícula, todos os grupos de dentes do sexo masculino obtiveram mais túbulos dentinários por aérea de quadrícula. Houve uma obstrução dos túbulos com o avanço da idade, no entanto, a comparação por contagem obteve resultados satisfatórios quando observados cada grupo separadamente, bem como a subdivisão das amostras por sexo. O mesmo ocorreu quando medido o tamanho dos túbulos dentinários. Os dentes decíduos possuem túbulos menores em relação aos permanentes, uma vez que o tamanho do túbulo é diretamente proporcional ao tamanho do dente. Entre todos os grupos os homens obtiveram diâmetro médio de túbulos maiores que os das mulheres, revelando para as amostras do sexo feminino uma maior taxa de preenchimento. Os resultados obtidos por EDS revelaram que há uma relação negativa entre os elementos Ca e P e Mg e C, refletindo para o esmalte um aumento em Ca e P - em relação à dentina - e concentrações de C e Mg maiores na dentina em relação ao esmalte. Os resultados de microssonda eletrônica mostraram com clareza a diferença química entre dentina e esmalte, para a dentina o teor de P2O5 varia de 28 a 36% e para o esmalte a variação foi de 39 a 41,5%. O MgO foi verificado em maiores concentrações na dentina, com valores de 1,2 a 1,7%; em contrapartida, para o esmalte a variação é entre 0,5 e 0,7%. O CaO teve concentrações de 49 a 51% no esmalte e para dentina de 34 a 44%. Nos gráficos de distribuição os elementos indicativos de idade foram SrO, K2O, Na2O e MgO no esmalte e o SrO, MgO, K2O, Na2O, SiO2 e CaO na dentina. O SrO foi apontado como o principal óxido envelhecedor, ou seja, aumenta com o avanço da idade. Esses resultados trazem uma visão inovadora para a comunidade forense, sendo um grande passo para delimitar as técnicas analíticas que trazem algum indicador de idade e sexo de indivíduos e, assim, contribuir para a identificação de vítimas desaparecidas e deslinde de crimes. Abstract: The teeth are the most enduring organs of the human body, being formed by minerals of the apatite group, mainly Ca5 (PO4)3 OH, and there may be in their structure 4% to 6% carbonate (CO3) ²- anions. One of the aspects of research in Forensic Sciences is based on refining processes of identification of cadavers and in this context several techniques have been applied to the tooth in order to obtain inferences about the age of individuals. The average size of tooth crystallites by X-ray diffraction (XRD) is commonly employed, starting from the premise that there is a variation in the size of the average crystallites as a function of age. Other techniques have also been employed, although still in a little diffused way. This research was carried out on thirty-seven human teeth, from consenting donors, where the teeth were concentrated in three large groups: permanent third molars, permanent nonthird molars and deciduous. The teeth were analyzed by X - ray diffractometry, by scanning electron microscopy (SEM) and electron microprobe. In the XRD analyzes the diffractograms obtained for each tooth were grouped for a comparison between each type of tooth and for the same tooth type in different individuals; the results were refined by the Scherrer equation to obtain the average crystal size. In scanning electron microscopy, the teeth were analyzed in secondary electrons, where morphological aspects such as dentinal tubules were quantified by area of the grid and size of the mean diameters. In EDS, the elements Ca, P, Mg and C were mapped. The oxides P2O5, CaO, Na2O, K2O, MgO, SiO2, SO3, SrO, Al2O3 and the elements F and Cl were also analyzed in electron microprobe. According to the results, there is a negative relationship between crystallite size and age of individuals. Younger teeth (deciduous) present average crystallites around 50 nm, while older teeth have an average crystal size between 30 and 40 nm. The third molar teeth presented an average size higher than expected for permanent teeth (60nm). In the dentinal tubule counts by ridge area, all groups of male teeth obtained more dentinal tubules per quadrant air. There was an obstruction of the tubules with the advancement of age, however, the comparison by counting obtained satisfactory results when observed each group separately, as well as the subdivision of the samples by sex. The same occurred when measuring the size of the dentinal tubules. The deciduous teeth have smaller tubules in relation to the permanent ones, since the size of the tubule is directly proportional to the size of the tooth. Among all groups, men had a mean diameter of tubules larger than those of women, revealing a higher filling rate for the female samples. The results obtained by EDS revealed that there is a negative relationship between the Ca and P and Mg and C elements, reflecting to the enamel an increase in Ca and P - in relation to dentin - and higher concentrations of C and Mg in the dentin in relation to the enamel. The electron microprobe results clearly showed the chemical difference between dentin and enamel, for dentin the P2O5 content varies from 28 to 36% and for enamel the variation was from 39 to 41.5%. The MgO was verified in higher concentrations in the dentin, with values of 1.2 to 1.7%; in contrast, for the enamel the variation is between 0.5 and 0.7%. The CaO had concentrations of 49 to 51% in the enamel and for dentin of 34 to 44%. In the distribution graphs the elements indicative of age were SrO, K2O, Na2O and MgO in the enamel and SrO, MgO, K2O, Na2O, SiO2 and CaO in dentin. SrO was pointed as the main aging oxide, that is, it increases with the advancement of age. These results bring an innovative vision to the forensic community, a great step to delineate analytical techniques that bring some indicator of age and sex of individuals and thus contribute to the identification of missing victims and resolution of crimes.
