Identificação de fitohormônios no extrato com potencial biotecnológico da microalga Scenedesmus subspicatus

Resumo: A atividade agrícola representa uma das principais atividades econômicas do Brasil e, sendo assim, a demanda por fertilizantes é alta. Em paralelo, os biofertilizantes vêm ganhando espaço no mercado, por serem uma alternativa mais sustentável, econômica e ecologicamente amigável. Dentre os biofertilizantes disponíveis no mercado internacional, muitos utilizam macroalgas e mais recentemente alguns a base de microalgas foram desenvolvidos. Este trabalho teve como objetivo geral definir a estrutura química das moléculas responsáveis pela atividade bioestimulante vegetal do extrato aquoso obtido a partir da biomassa da microalga Scenedesmus subspicatus. A microalga foi cultivada em meio WC e meio WC modificado, sendo observado um maior crescimento quando utilizado o meio modificado. A biomassa liofilizada dos cultivos foi submetida a extração aquosa, sob agitação mecânica em temperatura ambiente (10%, 1 h), sendo o processo realizado duas vezes. A fração aquosa resultante da extração foi então dialisada em membrana de 1 kDa, gerando a fração E (eluída) e fração R (retida). A atividade bioestimulante vegetal em sementes de tomate (Solanum lycopersicum) foi avaliada para a fração E, nas concentrações de 0,5, 1,0, 1,5 e 2,0 g/L, tendo como controle negativo água. As sementes tratadas com a fração E apresentaram um maior comprimento, volume e área dos hipocótilos quando comparadas às sementes tratadas com água. A fração E também foi analisada por RMN de 13C o que permitiu a identificação de três glicosídeos: 6-sulfo-?-D-quinovopiranose-?-(1??1)-glicerol, ?-Dgalactopiranose-( 1??6)-?-D-galactopiranose-(1??1)-glicerol e ?-D-galactopiranose- (1??1)-glicerol. As frações EC, provenientes de cultivos com meio WC modificado, e ECB, provenientes de cultivo com meio WC em biorreator, foram analisadas por HPLCMS para investigar a existência de fitohormônios. Foram identificados os seguintes hormônios: ácido abscísico, ácido indol-3-acético, benziladenina, cinetina, dihidrozeatina e zeatina nas amostras EC e ácido giberélico GA3 e zeatina nas amostras ECB. Também foram analisadas as subfrações E1-E6 da fração E, que apresentaram ácido abscísico, ácido giberélico, benziladenina, benziladenosina, dihidrozeatina, cinetina, cinetina ribosídio, isopenteniladenina, zeatina e zeatina ribosídio. Acredita-se que o extrato aquoso tenha potencial para a formulação de um produto biofertilizante, por passar por um processo de extração pouco oneroso e que não apresenta risco ao meio ambiente, além de conter hormônios vegetais, responsáveis pela atividade bioestimulante vegetal. Abstract: The agricultural activity represents one of the main economical activities in Brazil, and, therefore, the demand for fertilizers is high. At the same time, the biofertilizers are gaining space in the market, since they are sustainable, economical and eco-friendly. Among the biofertilizers available in the market, a lot of them uses macroalgae and, more recently, some microalgae products were developed. This present work had as main general objective to determine the chemical structure of the substances responsible by the plant biostimulating activity from the aqueous extract, obtained from the microalgae Scenedesmus subspicatus biomass. The microalgae was cultivated in WC medium and modified WC medium, with a higher growth observed when using the modified WC. The freeze-dried biomass was submitted to two sequential aqueous extractions, under magnetic stirring at room temperature (10%, 1h). The aqueous fraction from the aqueous extraction was dialyzed in a 1 kDa membrane and yielded fraction E (eluted) and fraction R (retained). The fraction E plant biostimulating activity in tomato seeds (Solanum lycopersicum) was evaluated in the fraction E concentrations 0,5, 1,0, 1,5 and 2,0 g/L, and had water as negative control. The seeds treated with the fraction E had hypocotils with higher lengh, volume and hypocotil area when compared with the seeds treated with water. Fraction E was also analysed by 13C RMN, which resulted in the identification of three glycosides: 6-sulfo-?-D-quinovopyranose-?- (1??1)-glycerol, ?-D-galactopyranose-(1??6)-?-D-galactopyranose-(1??1)-glycerol e ?- D-galactopyranose-(1??1)-glycerol. The fractions EC (modified WC culture medium cultures), and ECB (WC culture medium cultured in bioreactor) were analyzed by HPLC-MS with the purpose of identify plant hormones. The following phytohormones were identified: abscisic acid, benziladenine, dihydrozeatin, kinetin, kinetin riboside, indol-3-acetic acid, isopentenyladenine, zeatin and zeatin riboside in the EC fractions, and gibberelic acid GA3 and zeatin in ECB fractions. The E1-E6 subfractions from fraction E were also analysed by HPLC-MS with the same purpose. Abscisic acid, benziladenine, dihydrozeatin, gibberelic acid GA3, kinetin, kinetin riboside, indol-3- acetic acid, isopentenyladenine, zeatin and zeatin riboside were found in the subfractions. It is believed that the aqueous extract has great potential for a biofertilizer, because of its low cost and eco-friendly extraction procedure and due to its plant hormone content, responsible for the plant biostimulating activity.
