Papel de vesículas extracelulares na modulação dos fenótipos de agressividade e resistência em modelo de glioblastoma humano

Resumo: Os gliomas apesar de apresentarem baixa incidência (2% dos tumores malignos em geral) possuem altas taxas de mortalidade. O quimioterápico padrão para esse tipo de tumor é a Temozolomida (TMZ). Dentre os gliomas, os astrocitomas são os mais comuns e são classificados de acordo com o grau de agressividade (I a IV), sendo o IV conhecido como glioblastoma - GB. Apesar do progresso na terapia, o perfil de agressividade e os mecanismos de resistência tumoral dificultam o tratamento. Células de GB secretam vesículas extracelulares (VEs) e o conteúdo dessas vesículas pode estar associado à modulação de fenótipos como agressividade e resistência. Diversos alvos moleculares caracterizados na biologia tumoral vêm sendo descritos no conteúdo vesicular. No presente trabalho, um de nossos principais alvos é o gene supressor de tumor RECK e seu transcrito alternativo RECK-var 3 (associado a mau prognóstico) com o objetivo de avaliar o papel de ambos na modulação dos fenótipos de agressividade e resistência, via VEs, em modelo de GB humano. Foram utilizadas as linhagens A172, T98G, U87MG, U87MG-EGFR, U87MG-RECK e U87MG-RECK var 3, diferentes em relação ao grau de agressividade e nível de expressão endógena dos transcritos. Nossos resultados mostram que VEs derivadas destas células foram caracterizadas como uma população majoritariamente de microvesículas (cerca de 200 nm). A internalização das VEs foi observada na linhagem A172 incubada com VEs derivadas das outras linhagens. Houve aumento na proliferação e na capacidade de formação de colônias em substrato sólido das células A172 após incubação com VEs de células mais agressivas (U87MG, U87MG-EGFP, U87MG-RECK var 3), sugerindo que essas VEs modularam o fenótipo de agressividade das células A172. Ainda, os resultados sugerem um aumento da proteína RECK endógena nas células A172 após incubação com VEs derivadas da linhagem U87MG-RECK, assim como um aumento nos níveis de transcrito de RECK-var 3 após incubação com VEs da linhagem U87MG-RECK var3. Juntos, esses resultados mostram a participação de ambos os transcritos na modulação do fenótipo de agressividade. Comparando os perfis das células U87MG e T98G na responsividade à TMZ, foi observado que células U87MG incubadas com VEs derivadas de T98G (resistente à TMZ) e posteriormente tratadas com TMZ aumentaram a sua resistência ao quimioterápico. Por outro lado, VEs derivadas de U87MG incubadas em células T98G parecem sensibilizá-las à TMZ enquanto VEs derivadas de U87MG-RECK var 3 parecem reverter esse efeito, não alterando a resistência ao tratamento. Os resultados encontrados até o momento evidenciam que células de glioblastoma com níveis diferenciados de RECK e RECKvar 3 são capazes de modular o fenótipo proliferativo e de resistência tumoral via sua capacidade de secreção de VEs com características distintas. Abstract: Despite gliomas have a low incidence (2% of malignant tumors in general), they have high mortality rates. The standard chemotherapy for this type of tumor is Temozolomide (TMZ). Among gliomas, astrocytomas are the most common and are classified according to the degree of aggression (I to IV), the IV being known as glioblastoma - GB. Despite progress in therapy, the profile of aggressiveness and the mechanisms of tumor resistance make treatment difficult. GB cells secrete extracellular vesicles (EVs), and the contents of these vesicles may be associated with the modulation of phenotypes such as aggressiveness and resistance. Several molecular targets characterized in tumor biology have been described in the vesicular content. In the present study, one of our targets is the RECK tumor suppressor gene and its alternative RECK-var 3 transcript (associated with poor prognosis) to evaluate the role of both in modulating the phenotypes of aggression and resistance, via EVs, in a human GB model. The cell lines A172, T98G, U87MG, U87MG-EGFR, U87MG-RECK and U87MG-RECK var 3 were used, different in relation to the degree of aggressiveness and level of endogenous expression of the transcripts. Our results show that EVs derived from these cells were characterized as a population mainly of microvesicles (about 200 nm). The internalization of EVs was observed in the A172 cells incubated with EVs derived from the other cells. There was modulation in the proliferation and colony formation capacity on solid substrate of A172 cells after incubation with EVs of more aggressive cells (U87MG, U87MGEGFP, U87MG-RECK var 3), suggesting that these EVs modulated the cell aggressiveness phenotype A172. Furthermore, we can observe the modulation of the endogenous RECK protein in A172 cells after incubation with EVs derived from the U87MG-RECK cells, as well as an increase in the transcript levels of RECK-var 3 after incubation with EVs of the U87MG-RECK var3 cells. Together, these results suggest the participation of both transcripts in the modulation of the aggressiveness phenotype. Comparing the profiles of U87MG and T98G cells in response to TMZ, it was observed that U87MG cells incubated with EVs derived from T98G (resistant to TMZ) and subsequently treated with TMZ increased their resistance to chemotherapy. On the other hand, EVs derived from U87MG incubated in T98G cells appear to sensitize them to TMZ while EVs derived from U87MG-RECK var 3 seem to reverse this effect, without changing the resistance to treatment. The results found so far show that glioblastoma cells with differentiated levels of RECK and RECK-var 3 can modulate the proliferative and tumor resistance phenotype, mediated by the secretion capacity of EVs with different characteristics.
