Otimização de protocolos de embriogênese somática de cana-de-açúcar das variedades RB036066, RB036088 e RB036091 e transformação genética para tolerância ao estresse hídrico

Resumo: O estresse hídrico é uma das causas da queda na produtividade da cana-deaçúcar e impede a expansão da cultura para áreas com chuvas irregulares e sem irrigação. Para mitigar os danos causados pelo estresse, a transformação genética pode ser uma grande aliada, pois viabiliza a inserção de um gene externo no DNA vegetal. O gene P5CS de Vigna aconitifolia tem sido inserido em diferentes espécies para incremento no teor de prolina visando o efeito osmoprotetor e osmoregulador. Assim, o objetivo deste trabalho foi a integração do plasmídeo pJS107, contendo o gene P5CS e o gene bar como gene de seleção, em cana-de-açúcar por biobalística. Para isso, foram desenvolvidos protocolos de embiogênese somática para as variedades RB036088, RB036066 e RB036091. Foram realizados 5 tratamentos com concentrações de 2,4-D (0, 1, 3, 9, 18 ?M) para indução de calos e 3 tipos de explantes (porção apical, mediana e basal do segmento meristemático de folhas imaturas). A multiplicação dos calos foi realizada no mesmo meio de cultura da indução e a conversão em meio de cultura MS sem adição de reguladores. A multiplicação das plantas foi realizada utilizando meio MS com adição de 0,88 ?M de BAP e 0,46 ?M de KIN e no enraizamento testou-se os meios MS, MS com metade dos sais, MS com adição de 70 g L-1 de sacarose e MS acrescido de 2,46 ?M AIB, também foi avaliada a taxa de sobrevivência na aclimatização. As concentrações mais recomendadas para indução e posterior conversão de calos foi de 3 ?M de 2,4-D para as variedades RB036091, RB036066 e 9 ?M de 2,4-D RB036088. Para o enraizamento e sobrevivência na aclimatização o meio MS com 70g L-1 de sacarose para as variedades RB036088 e RB036091 e o MS meia força para a variedade RB036066 foram os mais adequados. Com os resultados, a variedade RB036088 foi escolhida para ser utilizada nos testes de transformação. Por meio da biobalística, embriões somáticos foram transformados, com micropartículas de tungstênio e pressão de ruptura de 1.200 psi. A eficiência da transformação, considerando os putativos selecionados em meio seletivo, foi de 3%. Foram obtidos eventos, confirmados por PCR pela amplificação gene de seleção bar. Porém, eles não apresentaram sinal de expressão do gene bar na PCR em tempo real e não houve biossíntese de prolina superior ao controle negativo quando os eventos foram expostos, in vitro, a situação de estresse (-0,9 Mpa). O método utilizado para transformação dos embriões foi eficiente, porém o conteúdo de prolina nos eventos foi inferior ao esperado. Abstract: The drought stress is one the cause for low productivity in sugarcane and prevent the culture expansion for regions with irregular rainfalls and water limited environment. To mitigate damage caused by drought stress, the genetic transformation can be a great ally, because it viabilize the insertion of an external gene into plant DNA. The P5CS gene of Vigna aconitifolia has been inserted in different species to increase the proline content, aiming at the osmoprotective and osmoregulatory effect. Therefore, the objective of this work was the integration of pJS107 plasmid, containing the P5CS gene and the bar gene as selection gene, in sugarcane by biolistic. For that, they was developed somatic embryogenesis protocols for cultivars RB036088, RB036066 e RB036091. Five treatments with different concentrations of 2.4-D (0, 1, 3, 9, 18 ?M) for callus induction and 3 types of explants (apical, median and basal portion of the meristematic segment of immatture leaves) were performed. The multiplication of callus was performed in the same culture medium of induction and the conversion on MS culture medium without addition of regulators. The multiplication of plants was performed using MS medium with addition of 0.88 ?M of BAP and 0.46 ?M of KIN and it was tested for the rooting, MS medium, MS with half of the salts, MS with addition of 70 g L-1 of sucrose and MS plus 2.46 ?M of AIB, the survival rate in acclimatization was also evaluated. The most recommended concentrations for induction and subsequent callus conversion were 3 ?M of 2.4-D for cultivars RB036091, RB036066 and 9 ?M of 2.4-D for RB036088. For rooting and survival in acclimatization, the MS medium with 70 g L-1 sucrose for cultivars RB036088 and RB036091 and half-strength MS for cultivar RB036066 were the most suitable. With these results, the variety RB036088 was chosen to be used in the transformation tests. Through biolistic, somatic embryo were transformed, with tungsten microparticles and burst pressure of 1,200 psi. The transformation efficiency, considering the putatives selected in selective medium, was 3%. They were obtained events, confirmed by PCR through selection gene bar amplification. However, they do not presented the gene bar expression signal on real time PCR and there was not proline biosynthesis superior of negative control when the events were exposed, in vitro, to stress situation (-0.9 Mpa). The method used for embryo transformation was efficient, however the quantity of proline in the events was less than expected.
