Organogênese in vitro a partir de explantes foliares em três cultivares de mirtileiro

Resumo: O mirtileiro é uma espécie de clima temperado, que possui grande potencial de desenvolvimento no Brasil, principalmente na região sul do país, em vista das condições edafoclimáticas favoráveis. A produção de mudas desta espécie é maioritariamente feita de forma vegetativa. No entanto os métodos convencionais não apresentam a produção necessária para suprir o mercado interno, nem para promover o fomento da cultura. Uma das opções para a produção é a técnica de micropropagação, que em curto período de tempo e espaço reduzido, consegue produzir grande número de mudas. Desta forma, a elaboração de um protocolo de indução à organogênese até a formação de mudas é um fator essencial para a produção de mudas eficiente, especialmente para cultivares adaptadas às condições climáticas do sul do Brasil, tendo em vista que a maior parte dos estudos foi realizado com cultivares utilizadas no hemisfério norte. Desta forma, a presente pesquisa avaliou a resposta organogênica de explantes foliares não meristemáticos de três cultivares de mirtileiro (Bluegem, Climax e Delite) do grupo Rabbiteye tratados com os reguladores vegetais Thidiazuron (TDZ), 6-(y-y-dimethylallylamino)-purine (2iP), 6-benzylaminopurine (BAP) e Zeatina (ZEA) nas concentrações 0; 0,5; 2,5; 5; 10; 20 microM em meio de cultura Woody Plant Medium (WPM). Também foi realizada a etapa de multiplicação e aclimatização com enraizamento de mini estacas, utilizando o regulador vegetal ácido indol-3-butírico (AIB) nas concentrações 0, 250 e 500 mg L-1. A indução da organogênese nas cultivares Bluegem e Delite foi possível com a utilização do regulador TDZ na concentração 0,5 microM. Na cultivar Climax a organogênese foi observada com os reguladores vegetais TDZ e ZEA nas concentrações 0,5 e 10 microM. Em nenhuma das três cultivares foram observados resultados significativos com os reguladores 2iP e BAP. A multiplicação foi possível com a utilização do regulador vegetal ZEA na concentração 2,5 microM, sendo que esta promoveu o alongamento das brotações obtidas via organogênese. Para a aclimatização e enraizamento de mini estacas, o uso do regulador vegetal foi indiferente para as três cultivares avaliadas, cujo enraizamento de mini estacas e sobrevivência foi de 29,16% para Bluegem, 68,66% para Delite e 75% para Climax. Abstract: The blueberry is a species of temperate climate, which has great potential for development in Brazil, mainly in the southern region of the country, in view of the favorable edaphoclimatic conditions. The production of seedlings of this species is mostly done vegetatively. However, conventional methods do not have the necessary production to supply the internal market, nor to promote the culture. One of the options for production is the micropropagation technique, which in a short period of time and reduced space, manages to produce a large number of seedlings. In this way, the elaboration of an organogenesis induction protocol until the formation of seedlings is an essential factor for the production of efficient seedlings, especially for cultivars adapted to the climatic conditions of southern Brazil, considering that most of the studies are in cultivars used in the northern hemisphere. Thus, the present research evaluated the organogenic response of non-meristematic leaf explants of three blueberry cultivars (Bluegem, Climax and Delite) from the Rabbiteye group treated with the plant regulators Thidiazuron (TDZ), 6-(y-y-dimethylallylamino)- purine (2iP), 6-benzylaminopurine (BAP) and Zeatin (ZEA) at concentrations 0; 0.5; 2.5; 5; 10; 20 microM in Woody Plant Medium (WPM). The multiplication and acclimatization stage with rooting of mini cuttings was also carried out, using the plant regulator indole-3-butyric acid (IBA) at concentrations of 0, 250 and 500 mg L-1. The induction of organogenesis in Bluegem and Delite cultivars was possible with the use of the TDZ regulator at a concentration of 0.5 microM. In the Climax cultivar, organogenesis was observed with plant regulators TDZ and ZEA at concentrations of 0.5 and 10 microM. In none of the three cultivars, significant results were observed with the 2iP and BAP regulators. Multiplication was possible with the use of the plant regulator ZEA at a concentration of 2.5 microM, which promoted the elongation of shoots obtained via organogenesis. For the acclimatization and rooting of mini-cuttings, the use of the plant regulator was indifferent for the three evaluated cultivars, which rooting of cuttings and survival was 29.16% for Bluegem, 68.66% for Delite, and 75% for Climax
