Estudo ultraestrutural comparativo do glicocálix em células absortivas intestinais de camundongo

Abstract

Resumo: Este trabalho foi feito com o objetivo de comparar o glicocálix de células absortivas intestinais de camundongos, de diferentes idades, a nível ultraestrutural. A procura de possíveis diferenças morfológicas na estrutura, no recém-nascido ou nas diversas etapas até a maturidade, deveu-se à busca de correlações entre elas e o fato de este tipo celular apresentar progressiva perda da sua capacidade de transportar macromoléculas proteicas – as imunoglobulinas entre elas – à circulação. Levantou-se a possibilidade de que o glicocálix fosse o componente celular a agir como barreira a estas moléculas, à medida que se fosse elaborando com o desenvolvimento do animal. O material colhido para exame constituiu-se de segmentos do terço médio do intestino delgado de camundongos de 0, 1, 8, 15 e 22 dias, fixados em glutaraldeído e pós fixados em tetróxido de ósmio, ambos com tampão cacodilato e adicionados de vermelho de rutênio. Com este marcador, observou-se um glicocálix proeminente, morfologicamente uniforme em todas as etapas examinadas. Além desta estrutura, foram observados diferentes tipos de vesículas, entre elas, nas três primeiras etapas, as recobertas, visíveis tanto na porção apical como nas laterais das células. Uma elaboração progressiva dos microfilamentos provenientes dos microvilos e da trama terminal ficou bem evidente com o avançar da maturidade dos animais. Estas observações nos levaram a concluir ser o glicocálix um dos elementos envolvidos na seleção do material a ser transportado e para tanto devendo apresentar

Summary: The aim of this work was to compare the ultra structure of the glycocalyx of intestinal absorptive cells from mice at different ages. We tried to find out whether any morphological difference, from the newborn to the adult, would explain the progressive loss of ability of these cells in the transport of intact proteins, including immunoglobulins, to the circulation. If the glycocalyx was the element acting as a barrier for these molecules in the adult, than it should be progressively more elaborated during the animal's postnatal development. Samples were colected from the small intestine of mice at the ages of 0, 1, 8, 15 and 22 days. Glutaraldehyde was used for fixation and osmium tetroxide for postfixation, both buffered with cacodylate and with the addition of ruthenium red. With the ruthenium red the glycocalyx showed conspicuous and ultrastructurally uniform in all the stages. Coated vesicles were evident in the cell apical region as well as nearby the lateral membranes at the first three stages examined. A progressive elaboration of the microfilaments coming from the microvilli and participating of the terminal web was verified with aging in the animals. These observations led us to conclude that the glycocalyx must be one of the elements involved in the selective uptake and transport of the molecules and therefore should progressively change the quality of its components since morphological changes cannot be detected. The other elements involved in the process must be the coated vesicles and membrane-associated cytoskeletal components.
