Germinação assimbiótica das microalgas na propagação in vitro de Cattleya labiata Lindl.

Resumo: Cattleya labiata Lindl. é uma orquídea nativa que está ameaçada de extinção. Para a sua preservação é necessário a formulação de protocolos que otimizem sua produção em larga escala. O objetivo desse estudo foi estabelecer um protocolo eficiente para o cultivo in vitro de C. labiata e avaliar o uso de microalgas Messastrum gracile e Chlorella vulgaris como substitutas de reguladores vegetais (BAP, TDZ e ZEA) na micropropagação utilizando a técnica "thin cell layer" (TCL). Os meios de cultura MS, MS/2 e WPM, com ou sem adição de 2 g L-1 de carvão ativado foram testados para a germinação in vitro. As plântulas obtidas foram subcultivadas para os mesmos meios para promover o alongamento e formação de raízes. O transplantio das mudas foi realizado testando diferentes substratos isolados ou combinados: fibra de coco (F), Sphagnum sp. (S), casca de Pinus sp. (C). As sementes cultivadas no meio MS/2C com os macronutrientes reduzidos pela metade e adição de carvão ativado, apresentaram maior porcentagem de germinação (71 %) e formação de plântulas (30 %), após 16 semanas de cultivo, em comparação com os outros meios de cultura (MS e WPM com ou sem carvão ativado). Na etapa de alongamento e desenvolvimento de raízes, os explantes cultivados em meio WPMC apresentaram maior comprimento da parte aérea e total (5,8 e 9 cm, respectivamente) em relação ao das cultivadas em outros meios, após 24 semanas de alongamento e após 24 semanas de transplantio (5,9 e 13 cm, respectivamente). Entretanto, as plantas cultivadas em meio WPMC apresentaram maior porcentagem de mortalidade (79,4 %). Recomenda-se o uso de MS/2C na germinação assimbiótica e no alongamento, pois as plantas cultivadas nesse meio apresentaram comprimento total e massa fresca elevados e alta porcentagem de sobrevivência, comparado com as cultivadas em outros meios. O substrato recomendado é o Sphagnum sp. Na técnica TCL, os protocormos obtidos da germinação in vitro foram testados inteiros ou secções transversais e longitudinais. Os explantes foram inoculados em meio MS/2 contendo extrato Messastrum gracile (EM), extrato de Chlorella vulgaris (EC), biomassa de Messastrum gracile (BM) ou biomassa de Chlorella vulgaris (BC), ou reguladores vegetais (BAP, TDZ e ZEA) em diferentes concentrações. Os explantes obtidos foram subcultivados para meio MS/2 com 2 g L-1 de carvão ativado e as plantas foram transplantadas em bandejas de semeadura contendo Sphagnum sp. como substrato. Os explantes cultivados em meio suplementando com BM e BAP apresentaram maior porcentagem de regeneração de PLBs (59 e 48 % respectivamente) e alto número médio (4 e 10 PLBs por explante, respectivamente). Na etapa de alongamento, as melhores respostas de comprimento total ocorreram com os tratamentos de adição de BM (7 cm) e BAP (6 cm) e também elevada porcentagem de sobrevivência no transplantio (BM: 85 % e BAP: 90 %). O TDZ não é recomendado, pois os explantes apresentaram menor comprimento no alongamento (4 cm), levando mais tempo para formação de raízes e menor porcentagem de sobrevivência no transplantio (40 %). A suplementação com as microalgas M. gracile e C. vulgaris pode substituir os reguladores vegetais, e favorecer a formação de PLBs e plantas. Para a propagação de C. labiata, recomenda-se a suplementação do meio com concentrações de 4 g L-1 de BM ou 4 mg L-1 de BAP. Abstract: Cattleya labiata Lindl. is an orchid that is in danger of extinction. Protocols are important to optimize their seedling production. The objective of this study was to establish an efficient protocol for in vitro cultivation of C. labiata and to evaluate the use of Messastrum gracile and Chlorella vulgaris microalgae as substitutes for regulators (BAP, TDZ and ZEA) in micropropagation using "thin cell layer" (TCL) technique. Asymbiotic germination was performed in MS, MS / 2 and WPM, with or without the addition of 2 g L-1 of activated charcoal. The plants underwent a period of elongation in the same medium as the germination and transplantation in different substrates isolated or combined: Coconut fiber (F), Sphagnum sp. (S), Pinus sp. (C). In the TCL technique, the sections were inoculated into the MS / 2 medium including M. gracile extract (EM), C. vulgaris extract (CE), M. gracile biomass (BM) or C. vulgaris biomass (BC), or regulators (BAP, TDZ and ZEA) at different concentrations. The explants underwent a period of elongation in MS / 2C with 2 g L-1 of activated charcoal and transplantation in Sphagnum sp. Explants cultivated in MS / 2C medium with macronutrients reduced by half and addition of activated charcoal presented higher percentage of germination (71 %) and plant formation (30 %), after 16 weeks of cultivation, in comparison to other culture media (MS and WPM with or without activated charcoal). The plants cultivated in the WPMC presented total and aerial part length superior (5.8 and 9 cm respectively) in relation to those cultivated in other medium, after 24 weeks of elongation and (5.9 and 13 cm respectively) after 24 weeks of transplantation. However, the plants cultivated in the WPMC medium showed the highest percentage of mortality. The use of MS / 2C in asymbiotic germination and elongation is recommended, as the plants cultivated in this medium presented high total length and fresh mass and high percentages of survival, compared to those cultivated in other media. The recommended substrate is Sphagnum sp. In the TCL technique, explants cultivated in medium supplemented with BM presented higher percentage of PLBs regeneration (59 %) and a high average number (4 PLBs per explant). In elongation, they presented high total length (7 cm) and high percentage of survival in the transplantation (85 %). TDZ is not recommended, because presented shorter length in elongation (4 cm), taking longer to form roots and lower percentage of survival in transplantation (40 %). Supplementation with microalgae M. gracile and C. vulgaris can replace regulators and favors the formation of PLBs and plants. For the propagation of C. labiata, medium supplemented with high concentrations of BM (4 g L-1) or BAP (4 mg L-1) is recommended.
