Micromorfologia e anatomia da flor de espécies de Aspigogyne e microchlus (goodyerinae - orchidaceae)

Resumo: Orchidaceae é uma das maiores famílias de angiospermas, possui distribuição cosmopolita e encontra-se bem representada no território brasileiro, aparecendo em todos os tipos de formação vegetal. A família encontra-se subdividida em cinco subfamílias e, dentro da subfamília Orchidoideae, a tribo Cranichideae abrange cerca de 100 gêneros e cerca de 1.800 espécies predominantemente terrícolas, sendo subdividida em oito subtribos: Chloraeinae, Cranichidinae, Galleotiellinae, Goodyerinae, Manniellinae, Pterostylidinae, Discyphinae e Spiranthinae. Dessas, a subtribo Goodyerinae é representada no Brasil por 37 espécies nativas em dois gêneros: Aspidogyne Garay e Microchilus C. Presl. No estado do Paraná são conhecidas 14 espécies do gênero Aspidogyne e duas espécies do gênero Microchilus. Os gêneros desta subtribo possuem a formação de um calcar, originado do labelo, cuja morfologia tem importância taxonômica. Considerando a escassez de conhecimentos anatômicos e da micromorfologia floral desta subtribo, o propósito do trabalho foi caracterizar a estrutura anatômica e micromorfológica das peças florais de espécies de Aspidogyne, Microchilus e Ludisia discolor, com o objetivo de identificar e caracterizar possíveis estruturas secretoras nas peças florais e em especial no calcar, assim como o processo secretor. As análises foram realizadas por meio da microscopia de luz, microscopia eletrônica de varredura e transmissão seguindo técnicas convencionais em anatomia vegetal. Com exceção de A. bidentífera e A. kuczynskii, os tricomas foram encontrados na face abaxial das sépalas de todas as espécies, na face externa do ovário e em algumas espécies no interior do calcar. As papilas foram encontradas nas sépalas, labelo e calcar de todas as espécies, apresentando formato e localização diversificada, e não tendo necessariamente características de tecido secretor. Epiderme secretora foi encontrada na face abaxial das sépalas laterais apenas em A. bidentifera e na face abaxial do labelo de A. commelinoides e M. arietinus. O calcar é distinto entre as espécies, apresentando nectários característicos em seu interior. Observou se na fase pré-antese o tecido epidérmico e subepidérmico com intensa atividade metabólica, com muitos plastídios contendo grãos de amido, mitocôndrias, e vesículas que se juntam a plasmalema e liberam o seu conteúdo em uma região subcuticular, distendendo a cutícula. Em um próximo estágio da flor, na fase pós-antese, o conteúdo celular é bastante diminuto e a secreção que antes estava na região subcuticular é liberada para o lúmem do calcar através de poros na cutícula, não havendo rompimento da mesma. É notória também a grande proporção de floema em relação ao xilema nos feixes vasculares próximos aos nectários. Estes são circundados por células com características secretoras. Os dados analisados atestaram que o calcar é a estrutura nectarífera presente na flor, sendo a secreção do tipo granulócrina. Os resultados de micromorfologia e anatomia floral das espécies estudadas podem ser usados como subsídio para futuros estudos ecológico, buscando uma melhor compreensão da interação entre essas plantas e seus polinizadores. Abstract: Orchidaceae is one of the largest families of angiosperms, has cosmopolitan distribution and is well represented in Brazilian territory, appearing in all types of plant formation. The family is subdivided into five subfamilies and within the subfamily Orchidoideae Endl., the Cranichideae tribe comprises about 100 genera and about 1,800 predominantly terrestrial species, being subdivided into eight subtribes: Chloraeinae, Cranichidinae, Galleotiellinae, Goodyerinae, Manniellinae, Pterostylidinae, Discyphinae and Spiranthinae. Of these, the subtribe Goodyerinae Klotzsch is represented in Brazil by 37 native species in two genera: Aspidogyne Garay and Microchilus C. Presl. In the state of Paraná 14 species of the genus Aspidogyne and two species of the genus Microchilus are known. The genera of this subtribe have the formation of a spur, originating from the labellum, whose morphology has taxonomic importance. Considering the scarcity of anatomical knowledge and the floral micromorphology of this subtribe, the purpose of the work was to characterize the anatomical and micromorphological structure of the floral parts of Aspidogyne, Microchilus and Ludisia discolor species, with the objective of identifying and characterizing possible secretory structures in flower parts in the spur, as well as the secretory process. The analyzes were performed using light microscopy, scanning electron microscopy and transmission following conventional techniques in plant anatomy. With the exception of A. bidentífera and A. kuczynskii, trichomes were found on the abaxial surface of the sepals of all species, on the outer surface of the ovary and some species within the spur. The papillae were found in the sepals, labellum and spur of all species, presenting a diverse format and location, and not necessarily having characteristics of secretory tissue. Secretory epidermis was found on the abaxial surface of the lateral sepals only in A. bidentifera and in the abaxial surface of the labellum of A. commelinoides and M. arietinus. The spur is distinct among the species, presenting characteristic nectaries in its interior. In the transmission electron microscopy analysis of this region, epidermal and subepidermal tissue with intense metabolic activity were observed in the pre-anthesis phase, with many plastids containing starch grains, mitochondria, and vesicles that join the plasmalema and release their content in a subcuticular region, relaxing the cuticle. In the next stage of the flower, in the post-anthesis phase, the cellular content is very small and the secretion that was previously in the subcuticular region is released to the lumen of the calcar through pores in the cuticle, and there is no rupture thereof. The large proportion of phloem in relation to the xylem in the vascular bundles near the nectars is also well known. These are surrounded by cells with secretory characteristics. The analyzed data attest that the calcar is the nectariferous structure present in the flower, being the secretion of granulócrina type. The results of micromorphology and floral anatomy of the studied species can be used as a subsidy for future ecological studies, seeking a better understanding of the interaction between these plants and their pollinators.
