Produção de compostos bioativos de Ganoderma lucidum por fermentação em estado sólido : avaliação da ação antitumoral, imunomoduladora e hipolipidemica

Resumo: Ganoderma lucidum é um cogumelo medicinal amplamente empregado no Oriente para o tratamento de diversas doenças. Neste trabalho, G. lucidum foi desenvolvido por cultivo submerso, testando-se diferentes variáveis físico-químicas, com o objetivo de se obter a produçao máxima de polissacarídeos. O micélio obtido por cultivo submerso foi desenvolvido por fermentaçao em estado sólido, empregando-se trigo em grao como substrato. A partir desse produto, foram preparadas as seguintes raçoes: G85 e G50, contendo 85% e 50% do fermentado de G. lucidum, respectivamente. Também foi testada uma raçao contendo 50% de fermentado de G. lucidum e 50% de fermentado de Agaricus brasiliensis (raçao A+G). Durante 14 semanas, foram alimentados 80 camundongos, divididos em quatro grupos de 20 indivíduos, denominados G85, G50, A+G e controle, em funçao da sua dieta. O grupo controle foi alimentado com raçao sem cogumelos. Na décima semana, os animais foram divididos em oito grupos constituídos de dez indivíduos, sendo que quatro grupos foram denominados: G85, G50, A+G e controle, os quais receberam injeçao interscapular de soluçao fisiológica. Os quatro grupos restantes foram denominados G85-S, G50-S, A+G-S e controle-S, nos quais foram implantadas células de sarcoma 180 na regiao interscapular. Na 14S semana, foram avaliadas as propriedades imunomoduladora, antitumoral e hipolipidemica das raçoes. Foi observada regressao tumoral nos grupos G50-S (89%), A+G (81%) e G85-S (14%) em relaçao ao controle-S. Em relaçao ao grupo controle, nos animais injetados com salina foram observadas as seguintes alteraçoes em células esplenicas: G85 apresentou aumento de células CD19+ (56%); G50 apresentou aumento de CD19+ (134%) e diminuiçao de CD4+ (-15%); enquanto que A+G apresentou elevaçao de células CD3+ e CD8+ (32,4 % e 34,6%, respectivamente). Em relaçao ao grupo controle-S, nos animais injetados com sarcoma 180 foram observadas as seguintes alteraçoes em células esplenicas: CD3+ aumentou 49,3%, 86% e 112,4% nos grupos G85-S, G50-S e A+G-S, respectivamente; CD4+ aumentou 38%, 56,4% e 68,7% nos grupos G85-S, G50-S e A+G-S, respectivamente; CD8+ aumentou 30,5%, 41,2% e 49% nos grupos G85-S, G50-S e A+G-S, respectivamente. Nos animais inoculados com salina, apenas o grupo G50 apresentou concentraçoes detectáveis de IFN-., além de aumentar em 16% as concentraçoes plasmáticas de TNF-a, em relaçao ao controle. Os grupos G85 e A+G nao apresentaram alteraçoes significativas nas concentraçoes de interleucinas, em relaçao ao grupo controle. Na presença do tumor, os grupos G85-S, A+G-S e controle-S apresentaram elevaçao nos níveis plasmáticos de IFN-. e TNF-a, enquanto que no grupo G50-S as concentraçoes dessas interleucinas estavam diminuídas. O consumo da dieta suplementada com G. lucidum causou significativas alteraçoes no metabolismo lipídico, particularmente a raçao G50, a qual levou a um decréscimo nos níveis plasmáticos de colesterol (-17%) e triacilgliceróis (-42%), além de aumentar 127% a fraçao HDL-colesterol. Esses resultados sugerem o possível emprego de G. lucidum como fonte para o desenvolvimento de novos fármacos e substâncias nutraceuticas com propriedades imunomoduladora, antitumoral e hipolipidemica. Abstract: Ganoderma lucidum is a medicinal mushroom widely used in the Far East for treatment of several diseases. In the present work, G. lucidum was grown by submerse cultivation, and different physical-chemical variables were tested in order that the maximal polysaccharides production could be obtained. Mycelia from submerse cultivation was grown by solid-state fermentation, employing wheat grain as substrate. From this product, the following diets were prepared: G85 and G50, containing 85% and 50% of the G. lucidum fermented product, respectively. A diet containing 50% of the G. lucidum fermented product and 50% of the Agaricus brasiliensis fermented product (A+G diet) was also tested. During 14 weeks, 80 mice were fed on it. The animals were divided into four groups of 20 animals each and labeled according to the diet they were taking: G85, G50, A+G and control group. The control group was fed a diet without mushroom. In the 10th week, the animals were divided into eight groups consisting of 10 individuals each, and four of these groups were labeled G85, G50, A+G, and control, and these received interscapular injection of a physiological solution. The four remaining groups were labeled G85-S, G50-S, A+G-S, and control-S, and these had sarcoma 180 cells implanted in their interscapular region. In the 14th week, the immunomodulatory, antitumoral and hypolipidemic properties of these diets were assessed. Tumoral regression was detected in the G50-S (89%), A+G-S (81%), and G85-S (14%) groups, as compared to the control-S. In relation to the control group, were detected the following alterations in the spleenic cells from animals injected with the saline solution: G85 showed an increase of CD19+ cells (56%); G50 showed an increase of CD19+ cells (134%) and a decrease of CD4+ cells (-15%); A+G group showed an increase of CD3+ and CD8+ cells (32.4% and 34.6%, respectively). In relation to the control-S group, were detected the following alterations in the spleenic cells from animals injected with sarcoma 180: CD3+ increased 49.3%, 86%, and 112.4% in groups G85-S, G50-S and A+G-S, respectively; CD4+ increased 38%, 56.4%, and 68.7% in groups G85-S, G50-S, and A+G-S, respectively; CD8+ increased 30.5%, 41.2%, and 49% in groups G85-S, G50-S, and A+G-S, respectively. Among the healthy animals, only G50 group showed detectable concentrations of IFN-?, besides a 16% increase of plasmatic concentrations of TNF-a, when compared to the control group. Groups G85 and A+G did not show any significant alterations in the cytokine concentrations. In the presence of the tumor, groups G85-S, A+G-S, and control-S showed an increase in the plasmatic concentrations of IFN-? and TNF-a, whereas in the G50-S group the concentration of these cytokines decreased. Diet supplemented with G. Lucidum resulted in significant alterations in the lipidic metabolism, specially the G50 diet, which led to a decrease in the plasmatic concentrations of cholesterol (-17%) and triacylglycerols (-42%), besides producing a 127% increase in the HDL-cholesterol. These results point at the possibility of using G. lucidum as source for developing new medicinal drugs and nutraceutic substances with immunomodulador, antitumoral, and hypolipidemic properties.
