Phytochemical evaluation, antioxidant activity and propagation methods of Baccharis dracunculifolia D.C : the main botanical source of brazilian green propolis

Resumo: A espécie Baccharis dracunculifolia DC., conhecida popularmente como "alecrim-do-campo", é um arbusto dióico nativo do Brasil com grande importância econômica devido aos seus metabólitos secundários. A planta é conhecida por ser a principal fonte botânica de um importante produto comercial conhecido como "própolis verde", que possui várias propriedades benéficas para a saúde humana, entre elas propriedades antitumorais. O metabolismo secundário vegetal é influenciado por fatores genéticos e ambientais, e o entendimento de como esses fatores influenciam a diversidade química em populações naturais é o primeiro passo para satisfazer com sucesso as demandas industriais, sendo possível, posteriormente, selecionar acessos altamente produtivos para cultivos e programas de domesticação. Neste sentido, o objetivo principal deste trabalho foi avaliar a variabilidade química dos metabólitos secundários das folhas de B. dracunculifolia e suas atividades antioxidantes em populações de ocorrência natural no sul do Brasil, além de avaliar as técnicas de propagação da espécie. Os extratos vegetais foram analisados por HPLC-DAD e LC-MS, a análise da composição do óleo essencial (OE) foi realizada por GC-FID e GC-MS e as atividades antioxidantes foram analisadas pelos métodos DPPH e ABTS. Além disso, foram avaliadas técnicas de propagação (1) por reprodução sexual: curva de maturação para determinar o ponto de maturidade fisiológica e definir melhores condições (temperatura) para a germinação; (2) por reprodução assexuada: a influência do sexo da planta matriz no enraizamento de estacas de caule de acordo com diferentes concentrações de ácido indolbutírico. As populações naturais estudadas produziram diferentes teores de óleo essencial e distintas quantidades de compostos terpênicos, e isso pode estar relacionado principalmente ao fator genético e secundariamente aos fatores bióticos e abióticos a que as plantas foram expostas, o que também teve reflexo nas atividades antioxidantes apresentadas. Apesar da diferença quantitativa nos compostos, a composição química do óleo essencial não foi alterada. As populações naturais estudadas podem ser divididas em três grupos principais, de acordo com os níveis de seus compostos terpênicos. Ao longo do ano existem variações no teor, na quantidade dos compostos e na atividade antioxidante do óleo essencial, que podem ser divididos em duas épocas: I) verão e outono, fase de floração, onde o OE produz maior porcentagem de (EJ-nerolidol; II) inverno e primavera, fase vegetativa, OE produz maior porcentagem de B-pineno. O metabolismo vegetal de B. dracunculifolia é afetado tanto pelo gênero da planta e quanto pelo local de crescimento. As plantas masculinas produzem níveis mais altos de compostos fenólicos, assim como maior teor de óleo essencial e maior capacidade antioxidante. O artepillin C, composto mais importante para a comercialização da própolis verde, apresenta maior conteúdo nas folhas de plantas masculinas do que nas femininas. A maturidade fisiológica das sementes ocorre aos 40 dias após a antese e as temperaturas de 25°C e 20-30°C são as melhores para a germinação da espécie. A aplicação do IBA é essencial para o enraizamento de estacas, mas mesmo assim o enraizamento é baixo e não foi influenciado pelo gênero da planta matriz. Este é o primeiro relato de propagação vegetativa de B. dracunculifolia, entretanto estudos adicionais são necessários para otimização da metodologia de propagação da espécie. Abstract: Baccharis dracunculifolia DC., which widely known as "alecrim-do-campo" is a dioecious shrub native to Brazil. This plant has great economical importance due to its secondary metabolites. This species is popular for being the botanical source of the important commercial product "green propolis", which has several beneficial properties for human health, such as anti-tumor activity. Secondary plant metabolism is influenced by genetic and environmental factors. Thus, understanding how these factors influence chemical diversity in wild populations is the first step in successfully satisfying industrial demands, by being able to select highly productive accessions for crops and improvement of domestication. In this sense, the main objective of this study was to evaluate the chemical variability of secondary metabolites of B. dracunculifolia, and their antioxidant activities of wild populations in southern Brazil (Parana state) and also to assess its propagation techniques. The plant extracts were analyzed by HPLC-DAD and LC-MS, the essential oil composition was determined by GC-FID and GC-MS and their antioxidant activities were analyzed by DPPH and ABTS methods. In addition, propagation techniques was evaluated (1) by sexual reproduction: maturation curve to determine the physiological maturity point and define better condition (temperature) for germination; (2) by asexual reproduction: the influence of the plant sex on rooting of stem cuttings with different concentrations of indolbutyric acid (IBA). The wild populations reported to produce different essential oil contents and different amounts of terpenic compounds. This may be related mainly to the genetic factor and secondarily to the biotic and abiotic factors that the plants were exposed. It also had an impact on the antioxidant activities presented. The essential oil shows quantitative changes in its compounds, however, the chemical composition remained unchanged. In relation to the results, this population can be divided into three main groups, according to the levels of their terpenic compounds. Throughout the year, there were variations in the content, quantity of the compounds and antioxidant activity of the essential oils, which can be divided into two seasons: I) summer and autumn, flowering phase, EO produces a higher percentage of (EJ-nerolidol; II) winter and spring, vegetative phase, EO produces a higher percentage of B-pinene. The secondary metabolism of B. dracunculifolia is affected both by the plant's gender and by the growth site. Male plants produced higher levels of phenolic compounds and essential oil contents and also showed greater antioxidant capacity compared with female plants. Artepillin C, the most important compound for the commercialization of green propolis, was present in higher content in the leaves of the male plant than that of female plant. Moreover, the physiological maturity of seeds occur at 40 days after anthesis which being the best time for seed collection in the field. The temperatures of 25°C and 20-30°C are the best for germination tests of the species. The application of IBA is essential for rooting of stem cuttings, but even so the rooting is low and was not influenced by the gender of the matrix plant. This is the first study report on vegetative propagation of B. dracunculifolia, however further work is required to optimize the propagation methodology of the plant.
