Propagação in vitro do híbrido interespecífico de videira 'Southern Home'

Resumo: A cultivar 'Southern Home' é um híbrido interespecífico complexo entre espécies do subgênero Muscadínea e Vitis vinifera L., o qual foi recentemente introduzido no Brasil e apresenta frutos com propriedades nutracêuticas, alto vigor, resistência a doenças e, devido ao formato de suas folhas, é muito usado na ornamentação de jardins, casas e etc. Com o aumento da demanda por mudas, a micropropagação torna-se a principal técnica de produção de mudas de qualidade, em um curto espaço de tempo e em um pequeno ambiente. Assim, o objetivo deste trabalho foi desenvolver um protocolo eficiente de micropropagação da videira 'Southern Home' para a rápida produção de mudas de qualidade. Para o estabelecimento in vitro, brotações foram coletadas e desinfestadas com fungicida Cercobim® (2,5 g L-1), etanol 70% e hipoclorito de sódio a 2,5%. Posteriormente estas brotações foram excisadas em segmentos nodais de 7 mm com uma gema e sem folhas e foram inoculados em cinco diferentes meios de cultura (MS, QL, WPM, WPM modificado e DSD1). Na fase de indução de brotação, testou-se os mesmos meios de cultura da fase anterior e a ação de 6-benzaminopurina (BAP) e zeatina (ZEA) nas concentrações (0; 1,0; 2,5 e 5,0 ?M) em meio de cultura WPM modificado. Em seguida, verificou-se a ação do ácido 3-indolacético (IAA), ácido naftalenoacético (NAA) e ácido indolbutírico (IBA) nas concentrações (0; 0,2; 0,4 e 0,8 ?M) no enraizamento in vitro e seu efeito na aclimatização ex vitro das plantas cultivadas in vitro. No estabelecimento in vitro da videira 'Southern Home' o meio de cultura WPM modificado permitiu maior sobrevivência dos explantes (75,00%), maior porcentagem de brotação (57,50%) e brotações de boa qualidade (nota 2,40). Na indução de brotação, o meio WPM proporcionou alta porcentagem de brotação em todos os subcultivos (98; 92 e 98%), maior comprimento de brotação (3,43; 5,44 e 7,86 cm) e maior número de gemas por brotação (5,16; 6,30 e 8,68). Em relação as citocininas testadas, a concentração de 1 ?M de ZEA proporcionou o maior comprimento e qualidade de brotação, maior número de gemas e menor aparecimento de sintomas de hiperhidricidade nas brotações, sendo que a ausência de citocininas no meio proporcionou baixa porcentagem de brotação em todos os subcultivos analisados (43,75 e 20%). Em se tratando do enraizamento in vitro, houve 95% de explantes enraizados no meio de cultura sem auxinas (IBA, IAA e NAA) e de 99,44% na presença das auxinas testadas. Já a sobrevivência das plantas foi de 100%, tanto na etapa de aclimatização ex vitro em laboratório como na aclimatização em casa de vegetação. Conclui que foi estabelecido um protocolo eficiente de micropropagação do híbrido de videira 'Southern Home', com a garantia de produção de mudas de qualidade e que a introdução in vitro desta videira deve ser realizada em meio WPM modificado, a indução de brotação em meio WPM acrescido de 1 ?M de ZEA e não se faz necessário o uso de auxinas para o enraizamento in vitro e para o sucesso ex vitro da videira 'Southern Home'. Abstract: The cultivar 'Southern Home' is a complex interspecific hybrid between species of the Muscadine subgenus and Vitis vinifera L. It was recently introduced in Brazil and presents fruits with nutraceutical properties, high vigor, and disease resistance. Also, due to the format of its leaves, it is widely used in the ornamentation of gardens, houses, etc. With the increasing demand for seedlings, micropropagation has become the main technique to produce quality seedlings, in a short period and a small environment. Thus, the objective of this work was to develop an efficient micropropagation protocol of the 'Southern Home' grapevine for the rapid production of quality seedlings. For in vitro establishment, shoots were collected and disinfected with Cercobim® (2.5 g L-1) fungicide, 70% ethanol, and 2.5% sodium hypochlorite. Subsequently, these sprouts were excised into 7 mm nodal segments with one bud and no leaves and were inoculated in five different culture media (MS, QL, WPM, modified WPM and DSD1). In the budding induction phase, testing used the same previous culture media and the action of 6-benzaminopurine (BAP) and zeatin (ZEA) at concentrations (0; 1.0; 2.5 and 5.0 ?M) in a modified WPM culture medium. Then, the action of 3-indol acetic acid (IAA), naphthaleneacetic acid (NAA), and indol butyric acid (IBA) at concentrations (0; 0.2; 0.4 and 0.8 ?M) was verified on in vitro rooting, and on ex vitro acclimatization of in vitro grown plants. Furthermore, during the establishment of the in vitro 'Southern Home' grapevine, the modified WPM culture medium allowed the highest survival of explants (75.00%), the highest percentage of sprouting (57.50%), and sprouts with good quality (score 2.40). In the sprouting induction phase, the WPM medium provided a high sprouting percentage in all subcultures (98; 92 and 98%), greater sprouting length (3.43; 5.44, and 7.86 cm), and the greater number of buds per sprout (5.16; 6.30 and 8.68). Regarding the cytokinins tested, the concentration of 1 ?M of ZEA provided the greatest length and sprout quality, greater number of buds, and fewer symptoms of hyperhidricity in the sprouts. Moreover, when in the absence of cytokinins in the medium, a low percentage of sprouting was observed in all the subcultures (43.75 and 20%). For in vitro rooting, there were 95% of rooted explants in the culture medium without auxins (IBA, IAA and NAA) and 99.44% in the presence of them. The survival of the plants was 100% for the ex vitro acclimatization in the laboratory and greenhouse stages. It concludes that, a micropropagation efficient protocol of the hybrid 'Southern Home' grapevine was established, guaranteeing quality seedlings production. And that, in vitro introduction of this grapevine should be done in modified WPM medium; the induction of sprouting in WPM medium plus 1 ?M of ZEA; and the use of auxins is not necessary for the in vitro rooting and for the ex vitro success of the 'Southern Home' grapevine.
