Aplicação de biomassa de microalga em sementes de soja para mitigação do estresse salino

Resumo: A soja é o principal grão oleaginoso produzido e consumido mundialmente, devido a sua alta produção de proteína e diversidade de produtos. Em condições salinas, os danos na semente causam alterações no metabolismo, reduzindo a germinação e crescimento das plântulas, além da nodulação. As microalgas apresentam importantes características estimulantes, mas seu uso ainda é pouco difundido para a promoção do crescimento vegetal. Objetivou-se avaliar o efeito do uso da biomassa da microalga Asterarcys quadricellulare, obtida em sistema de produção mixotrófico, como fonte de aminoácidos livres para aplicação em sementes, na germinação, no crescimento de plântulas e no desenvolvimento das plantas, em condições salinas. A bioatividade da biomassa foi identificada por meio de bioensaio, promovendo o aumento do volume de hipocótilos de plântulas de tomate. O experimento foi realizado com duas cultivares de soja (Lança e Zeus), em duas etapas: a primeira, realizada no Laboratório de Análise de Sementes-UFPR, para determinar as concentrações críticas de NaCl e as melhores doses de biomassa para serem utilizadas na segunda etapa, a qual foi realizada em casa de vegetação, no Laboratório de Biofertilizantes da Fazenda Experimental Canguiri-UFPR, Pinhais- PR. Primeiramente testaram-se diferentes níveis de salinidade (zero, 40, 60 e 80 mmol L-1 de NaCl) e concentrações da biomassa da microalga misturada com inoculante de Bradyrhizobium japonicum (zero; 1; 1,5; 2,0 e 2,5 mg L-1) aplicados às sementes, avaliando a germinação e crescimento inicial de plântulas em condições controladas. Empregou-se delineamento inteiramente casualizado, em esquema de fatorial triplo, realizando-se análise de variância e comparação de médias por Tukey. A germinação das sementes diminuiu com 60 mmol L-1 de NaCl e não teve diferença entre as cultivares; a aplicação via sementes de biomassa da microalga Asterarcys quadricellulare foi importante para a manutenção do poder germinativo das sementes em condição salina. A salinidade afetou negativamente o comprimento das plântulas e das raizes. A aplicação da biomassa de A. quadricellulare nas doses de 1,5 e 2,5 mg L-1 colaboraram para o aumento do comprimento das raizes e parte aérea das plântulas. A partir desses resultados, selecionou-se para o segundo experimento as doses da biomassa de A. quadricellulare em mistura com inoculante Bradyrhizobium japonicum: zero; 1,5 e 2,5 mg L-1, e as concentrações de zero; 60 e 80 mmol L-1 de NaCl. Em casa de vegetação, o uso da biomassa da microalga A. quadricellulare nas sementes, foi eficaz para mitigar os danos no desenvolvimento das plantas de soja, colaborando para o aumento da altura, área foliar e diâmetro dos nódulos em condições salinas, bem como no número de nódulos sem salinidade, sendo eficientes as doses de 1,5 e 2,5 mg L-1. A aplicação de A.quadricellulare estimulou o metabolismo de aminoácidos das plantas. O aumento da prolina e da atividade das enzimas antioxidantes foram importantes para mitigar os danos do estresse salino. Sendo assim, considera-se promissor o uso da microalga A.quadricellulare como fonte de aminoácidos livres para biofertilizantes, por apresentar bioatividade promovendo o crescimento e mitigando os efeitos do estresse salino em soja ao estimular o sistema antioxidante das plantas. Abstract: Soy is the main oilseed grain produced and consumed worldwide, due to its high protein production and product diversity. In saline conditions, damage to the seed causes changes in metabolism, germination and seedling growth, in addition to nodulation. Microalgae have stimulating characteristics, but their use is still not widespread for the promotion of plant growth. The objective was to evaluate the effect of using the biomass of the microalgae Asterarcys quadricellulare, obtained in a mixotrophic production system, as a source of free amino acids for application in seeds, germination, seedling growth and plant development, in saline conditions. The bioactivity of the biomass was identified by means of a bioassay, promoting an increase in the volume of hypocotyls from tomato seedlings. The experiment was carried out with two soybean cultivars (Lança and Zeus), in two stages: the first, performed at the Seed Analysis Laboratory-UFPR, to determine the critical NaCl rules and the best biomass doses to be used in the second stage, which was carried out in a greenhouse, at the Biofertilizer Laboratory of Fazenda Experimental Canguiri - UFPR, Pinhais-PR. Firstly, different salinity levels (zero, 40, 60 and 80 mmol L-1 of NaCl) and biomass concentrations of the microalgae mixed with inoculant of Bradyrhizobium japonicum (zero; 1; 1.5; 2.0 and 2, 5 mg L-1) applied to the seeds, evaluating the germination and initial growth of seedlings under controlled conditions. A completely randomized design was used, in a triple factorial scheme, with analysis of variance and comparison of means by Tukey. Seed germination decreased with 60 mmol L-1 of NaCl and there was no difference between cultivars; the application via biomass seeds of microalgae Asterarcys quadricellulare was important for maintaining the germinative power of seeds in saline condition. Salinity negatively affected the length of seedlings and roots. The application of A. quadricellulare biomass at doses of 1.5 and 2.5 mg L-1 helped to increase the length of the roots and aerial part of the seedlings. From these results, the doses of the biomass of A. quadricellulare were selected for the second experiment mixed with inoculant Bradyrhizobium japonicum: zero; 1.5 and 2.5 mg L-1, and concentrations of zero; 60 and 80 mmol L-1 of NaCl. In a greenhouse, the use of A. quadricellulare microalgae biomass in the seeds was effective in mitigating the damage in the development of soybean plants, contributing to the increase in height, leaf area and nodule diameter in saline conditions, as well as in number of nodules without salinity, with doses of 1.5 and 2.5 mg L-1 being efficient. The application of A.quadricellulare stimulated the metabolism of plant amino acids. The increase in proline and the activity of antioxidant enzymes were important to mitigate the damage from salt stress. Therefore, the use of the microalgae A.quadricellulare as a source of free amino acids for biofertilizers is considered promising, as it presents bioactivity promoting growth and mitigating the effects of salt stress in soybeans by stimulating the antioxidant system of plants.
