Embriogênese somática em jabuticabeira (Plinia cauliflora (Mart.) Kausel)

Resumo: A jabuticabeira (Plinia cauliflora) e uma arvore nativa e endemica pertencente a
familia Myrtaceae, com distribuicao nas regioes Sudeste e Sul do Brasil.
Devido a natureza recalcitrante de suas sementes e a falta de metodos
eficientes de propagacao vegetativa, ainda nao estao estabelecidos pomares
comerciais de jabuticabeira. A embriogenese somatica pode ser uma
alternativa para obtencao de grande numero de plantas em curto espaco de
tempo, em boas condicoes fitossanitarias. Com isso, o objetivo do presente
trabalho foi desenvolver um protocolo de embriogenese somatica a partir de
sementes maduras de jabuticabeira. Os frutos foram coletados na cidade de
Vitorino-PR e transportados ate o laboratorio da UFPR, em Curitiba. As
sementes foram desinfestadas em etanol 70% (1 min) e hipoclorito de sodio 5%
(10 min), seguido de lavagens em agua destilada esteril. Os dois cotiledones
foram separados e introduzidos individualmente, junto com eixos embrionarios,
em tubos de ensaio contendo meio de cultura. Para a inducao de massas pro.
embriogenicas (MPE) foram testadas tres concentracoes de acido 2,4-
diclorofenoxiacetico (2,4-D) (5, 10 ou 15 ƒÊM sem carvao ativado e tambem
diferentes concentracoes de 2,4-D (100, 200, 300 e 500 ƒÊM), com carvao
ativado (0,5; 1 ou 2 g.L-1). Para iniciar a maturacao dos embrioes somaticos, foi
testado carvao ativado (CA) (1 g.L-1) e polietilenoglicol (PEG 4000) (30, 60 e 90
g.L-1). Em seguida, as massas contendo embrioes somaticos foram transferidas
a varios meios, testando carvao ativado (1 g.L-1), PEG (30 g.L-1), GelzanR (2;
2,5 e 3 g.L-1), BAP (2, 4, 8 ƒÊM) e 0,5 ƒÊM de BAP+ GA3 (1 e 2 ƒÊM). Para a
inducao de MPE, o melhor resultado (82,5% dos explantes) foi no meio MS,
acrescido de 300 ƒÊM de 2,4-D e 1 g.L-1 de CA. Durante a fase I da maturacao,
o meio mais indicado para formacao de embrioes somaticos em estadio
torpedo foi MS acrescido de 30 g.L-1 de PEG 4000 (77,5% de formacao de
embrioes em estadio torpedo). Nao houve conversao dos embrioes somaticos
em plantulas. A formacao de embrioes somaticos foi possivel ate o estadio
cotiledonar (3,05%), formando embrioes anormais. Os estudos anatomicos
mostraram o desenvolvimento dos embrioes somaticos assincronicos e a
reacao das celulas ao 2,4-D, levando a morte celular.

Abstract: The jabuticabeira (Plinia cauliflora) is a native and endemic tree belonging to the family Myrtaceae, with distribution in the Southeastern and Southern regions of Brazil. Due to the recalcitrant nature of its seeds and the lack of efficient methods of vegetative propagation, commercial jaboticaba orchards have not yet been established. Somatic embryogenesis can be an alternative to obtain large numbers of plants in a short time, in good phytosanitary conditions. With this, the objective of the present work was to develop a somatic embryogenesis protocol from mature seeds of jabuticabeira. The fruits were collected in the city of Vitorino-PR and transported to the UFPR laboratory in Curitiba. The seeds were disinfested in 70% ethanol (1 min) and 5% sodium hypochlorite (10 min), followed by washing in autoclaved distilled water. The two cotyledons were separated and introduced individually, together with embryonic axes, into test tubes containing culture medium. For the induction of pro-embryogenic masses (PEM), three concentrations of 2,4- dichlorophenoxyacetic acid (2,4-D) were tested (5, 10 or 15 ?M) without activated charcoal (AC) and then different concentrations of 2,4-D (100, 200, 300 and 500 ?M) with AC (0.5, 1 or 2 gL-1) were used. To initiate maturation of somatic embryos, activated carbon (CA) (1 g.L-1) and polyethylene glycol (PEG 4000) (30, 60 and 90 g.L-1) were tested. Then, masses containing somatic embryos were transferred to various media by testing activated charcoal (1 gL- 1), PEG (30 gL-1), Gelzan® (2, 2.5 and 3 gL-1), BAP , 4.8 ?M) and 0.5 ?M BAP + GA3 (1 and 2 ?M). For the induction of MPE, the best result (82.5% of the explants) was in the MS medium, plus 300 ?M of 2,4-D and 1 g.L-1 of CA. During phase I maturation, the most indicated medium for the formation of somatic embryos in the torpedo stage was MS plus 30 g.L-1 of PEG 4000 (77.5% embryo formation in the torpedo stage). There was no conversion of somatic embryos into seedlings. Formation of somatic embryos was possible until the cotyledonary stage (3.05%), forming abnormal embryos. The anatomical studies showed the development of somatic embryos asynchronous and the reaction of the cells to 2,4-D, leading to cell death. KEY WORDS: 2,4-D, Micropropagation, Myrtaceae
