Abstract: Herbaspirillum seropedicae is a betaproteobacteria found in association with several crops and can promote plant growth and increase productivity. This effective stimulus of plant growth depends on an efficient colonization of the host plant. The association with the host begins with the bacterial attachment onto the root surface, followed by the colonization on points of secondary root emergency and penetration through epidermis discontinuities. A rapid occupation of intercellular spaces then occurs, proceeding with xylem and aerial portions colonization. However, the molecular nature of the colonization is not clearly understood. In these associations, the key event seems to be the rhizosphere colonization. Therefore, the identification and characterization of genic products involved in the plant colonization by plant growth promoting rhizobacteria (PGPRs) such as H. seropedicae has scientific importance and economic interest to potentiate its application. This way, this work describes that the H. seropedicae lipopolysaccharide act as anchoring molecule to radicular lectins during attachment and that the exopolysaccharide act as matrix in biofilm formation on abiotic surfaces. Other molecular factors were also identified by transcriptomic analyses of bacteria during maize colonization, discriminating the molecular adaptations of H. seropedicae when perceiving the environment, attaching onto root surface and surviving in the first stages of rhizosphere colonization.
