Hidrólise de triglicerídeos e síntese de éster de ácido graxo em sistema de micelas reversas

Resumo: 0 objetivo deste trabalho foi o de estudar os principais parâmetros que regem a cinética e a estabilidade da lipase de Thermomyces lanuginosa (Lipoiase 100 L, Novozyme) em reações de hidrólise de triglicerídeos e síntese de éster de cadeia longa, em sistema de micelas reversas de AOT [2-etil-hexil-sulfossuccinato de sódio, 100 mM] em isooctano. Em reações de hidrólise, foram determinados os efeitos do parâmetros Wo ([HfeOMAOT]), concentração de enzima, pH, temperatura e concentração do substrato. Verificou-se que a lipase de Thermomyces lanuginosa apresentou um perfil de atividade tipicamente na forma de sino, com atividade máxima (1092,4 U/mg) em Wo de 15. As demais condições ótimas para a atividade da enzima foram pH 8,0 e 37 °C, com atividade máxima de 2163,6 U/mg. O estudo do efeito da concentração dos substratos tríbutirina (10 a 170 mM) e trioleína (10 a 250 mM) mostrou que a enzima não apresenta um comportamento cinético de Michaelis-Menten, sendo inibida por excesso dos substratos. A atividade máxima de 1854,6 U/mg foi obtida a 120 mM de tríbutirina e de 2144,5 U/mg a 150 mM de trioleína. Nos ensaios de estabilidade da enzima frente à temperatura (30-60 °C), verificou-se que esta sofreu ativação quando incubada a 30 °C durante 30 minutos,, obtendo-se para as demais temperaturas tempos de meia-vida de 30, 20 e 10 min respectivamente a 37, 50 e 60 °C. Os efeitos dos parâmetros da reação e do sistema, como temperatura t, °C, pH, Wo, e razão molar (R.M), foram estudados através de um delineamento fatorial 24"1. As variáveis mais importantes foram o pH e a temperatura e a interação entre elas. As melhores condições para síntese do laurato de etila foram 30°C, pH 5,6; Wo 10; e R.M 5,0 com rendimento de 90% (60 min) e atividade específica de 220 U/mg. ABSTRACT: This thesis aimed to study the principal parameters that control the kinetics and stability of the lipase from Thermomyces lanuginosa (Lipolase 100 L, Novozyme) in reactions of triglycerides hydrolysis and esters synthesis with long-chain fatty acids, within reverse micelles of 100 mM AOT [bis (2-ethylhexyl) sulfosuccinate] in isooctane. The effects of the level of water in the system (Wo), enzyme concentration, pH, temperature and substrate concentration were determined for the hydrolysis reaction. The activity of Thermomyces lanuginosa lipase showed a typical bell curve against water content, with a maximum activity of 1092 U/mg at at Wo of 15. Other conditions for maximum activity were pH 8.0 and 37 °C, under which conditions the activity was 2164 U/mg. Against substrate concentrations of 10 to 170 mM tributyrin and 10 to 250 mM triolein the enzyme did not show Michaelis-Menten kinetics, with apparent substrate inhibition at high substrate concentrations. The maximum activities were 1855 U/mg at 120 mM tributyrin and 2144 U/mg at 150 mM triolein. In the tests of enzyme temperature stability over the range 30-60 °C, showed that the enzyme was activated when incubated for 30 minutes at 30 °C, but lost half of its activity within 30, 20 and 10 minutes for the temperatures of 40, 50 and 60 °C, respectively. The effects of temperature, pH, Wo and molar ratio were studied using a 24"1 factorial design. The most important variables were the pH and the temperature and the interaction between them. The best conditions for ethyl laurate synthesis were 30 °C, pH 5.6, a Wo of 10 and a molar ratio of 5.0 with a reaction extent of 90% in 60 min with a specific activity of 220 U/mg.
