Avaliação aguda dos efeitos toxicológicos da deltrametrina em uma espécie de peixe fluvial nativo : estudo bioquímico e imunotóxico

Resumo : A deltametrina (DM), um piretróide sintético, é largamente utilizada em cultivos no Brasil, na medicina veterinária, no controle de pestes domésticas e na saúde pública, principalmente para controle de vetores. De modo geral os organismos não alvos, como as espécies aquáticas são extremamente sensíveis aos piretróides. O objetivo deste estudo foi avaliar o efeito de doses subletais de deltametrina através da atividade enzimática da colinesterase (ChE), glutationa S-transferase (GST) e etoxiresorufina-O-deetilase (EROD), e da migração celular. Analisar também a produção de óxido nítrico (NO) através da estimulação de macrófagos "in vitro", em uma espécie de peixe nativo Ancistrus multispinis. Foram utilizados três grupos (controle, DM0,1 e DM0,3 mg.kg-1) para as análises enzimáticas e outros seis grupos com 10 peixes cada (controle, lipopolissacarídeo - LPS, LPS+DM0,1, LPS+DM0,3, DM0,1 e DM0,3 mg.kg-1, via IP) para a migração celular. As colheitas de sangue, exsudato peritoneal e tecidos (músculo, brânquias e fígado) foram realizadas 96 horas após a exposição ao piretróide. Procedeu-se à contagem total de leucócitos e de eritrócitos, taxa de hemoglobina e hematócrito sangüíneo, contagem total de leucócitos do exsudato peritoneal e análises enzimáticas. O cultivo celular foi realizado extraindo os macrófagos dos rins dos peixes, as células foram ajustadas a 2x106 cel.mL-1, incubado por 24 horas (23°C). Após este período foram realizados 6 tratamentos (meio, LPS 10µg.mL-1, LPS+DM0,3, LPS+ DM1, DM0,3 e DM1 µg.mL-1) por 48 horas, sendo realizado a dosagem de óxido nítrico. Os dados foram analisados por ANOVA seguidos por teste de Bonferroni. Houve aumento significativo da atividade colisterásica, redução significativa da GST e não houve alteração da EROD nos grupos com DM. Os resultados obtidos mostraram aumento significativo de todos os grupos tratados para os valores dos leucócitos e eritrócitos no sangue quando comparados com o grupo controle. Os valores da taxa de hemoglobina foram maiores nos grupos com DM na maior dose. Não houve alterações nos valores do hematócrito. Na cavidade peritoneal do A. multispinis existe naturalmente uma pequena população de leucócitos, porém após a inoculação de LPS a migração de leucócitos aumentou significativamente. No cultivo celular houve aumento significativo na quantidade de nitrito de todos os grupos tratados em relação ao controle. O grupo LPS + DM1µg.mL1 também apresentou um aumento em relação aos utros grupos tratados. Podemos concluir que a DM causou uma leucocitose em A. multispinis e que essa espécie é um bom modelo para avaliar efeitos tóxicos de piretróides e para estudos de migração celular. O método in vitro pode ser utilizado para avaliar efeitos potenciais de agentes tóxicos do sistema imune de peixe e, a DM mostrou ser um ativador de NO através dos macrófagos de A. multispinis tal qual o LPS. Abstract : Deltamethrin (DM), a synthetic pyrethroid, is widely applied in Brazilian crops, in the veterinary medicine, in the pest control and in public health against vectors. However, non-target species such as the aquatic organisms, mainly fish, DM is sensitive to pyrethroids. The aim of this study was to evaluate the effect of sublethal doses of DM through enzymatic activity of cholinesterase (ChE), gluthatione S-transferase (GST) and ethoxiresorufin-O-deethylase (EROD), and cellular migration. The nitric oxide (NO) was analysed through machophage stimulation "in vitro" from A. multispinis, a native fish. Three groups of animals were used (control, DM 0.1 and DM 0.3 mg.kg-1) for enzymatic analysis, and six other groups were used (control, LypopolissacharideoLPS, LPS+DM 0.1, LPS+DM0.3, DM 0.1 and DM 0.3 mg.kg-1, IP), each one with 10 fish for cellular migration. The blood and tissues (muscle, gills and liver) were collected 96 hours after the pyrethroid exposure. Total leukocytes and erythrocytes count, hemoglobin rate and hematocrit, leukocytes of peritoneal exsudate, enzymatic and histopathological analysis were carried out. Macrophages were collected from the fish kidney, the cells 2x106 cells.mL-1, incubated for 24 h at 23°C for adherence. Six treatments were carried out (medium, LPS 10µg.mL-1, LPS+DM0, 3, LPS+ DM1, DM0, 3 and DM1 µg.mL-1 ) for 48 h and the NO dosage analysed. The data were analyzed by ANOVA followed by Bonferroni's test. It was observed a significant increasing of the ChE activity and significant reduction of GST and no significant alteration of EROD in the DM groups. The blood leukocytes and erythrocytes of all the groups were significantly different to the control group. The hemoglobin levels increased in the highest dose. There was not alteration in the values of the hematocrit. There was naturally in the cavity peritoneal a small leukocytes population, however after inoculation of LPS the leukocytes migration increased significantly in relation to control animals. The NO production increased in all the treated groups in relation to control group. The group DM 1µg.mL-1 induced a significant increase in relation to other groups too. The DM caused a leucocytose in A. multispinis and this specie is a good model to evaluate toxic effects of pyrethroid and to the cellular migration studies. The "in vitro" method can be used to evaluate potential effects of toxic agents of fish immune system. DM could be an activator of NO production of A. multispinis as LPS.
