Imunosseleção de mimotopos de anticorpos bovinos contra Cysticercus bovis : desenvolvimento de um ensaio diagnóstico ante-mortem para cisticercose bovina

Resumo: A cisticercose é uma importante doença parasitária provocada pelo desenvolvimento dos metacestodeos de Taenia saginata (Cysticercus bovis) e de Taenia solium (Cysticercus celullosae) nos tecidos bovinos e suínos, respectivamente ou humanos. Além de causar prejuízos à pecuária de corte e ser considerada como um grande problema de saúde pública, ensaios diagnósticos sensíveis e precisos para detecção da parasitose no animal vivo e programas de vacinação permanecem como um desafio. Os objetivos deste estudo foram identificar, pela técnica de Phage Display, potenciais peptídeos ligantes a imunoglobulinas G de bovinos infectados experimentalmente com ovos de T. saginata, na tentativa de encontrar um bom antígeno para ser usado na detecção ante-mortem da cisticercose bovina e avaliar se um peptídeo sintético também identificado por Phage Display seria capaz de proteger animais contra infecção experimental. No primeiro capítulo desta dissertação estão apresentados os resultados referentes à cisticercose bovina e mostram que fagos obtidos diferenciam soros positivos e negativos para a parasitose. No capítulo 2, estão apresentados os resultados da avaliação da proteção de NC-1 em camundongos infectados experimentalmente com Taenia crassiceps, e demonstram que a proteção foi de 74%. Conclui-se que o Phage Display é uma técnica capaz de identificar peptídeos de interesse e estes são capazes de proteger animais em programas de imunização experimental. Abstract: Cysticercosis is an important parasitic disease caused by developing metacestodes of Taenia saginata (Cysticercus bovis) and Taenia solium (Cysticercus celullosae) in bovine and swine tissues, respectively or human. Besides the economic losses to beef cattle and its impact on the public health system, sensible and accurate diagnostic methods for the detection of the etiological agents of cysticercosis in infected animals, as well as the implementation of vaccination programmes remain a challenge. Thus, the present study aims to identify, through the Phage Display technology, potential ligand peptides interacting with imunoglobulins G of bovines experimentally infected with T. saginata eggs, in an attempt to find effective antigens to be used in the ante-mortem detection of bovine cysticercosis and to evaluate if a synthetic peptide also identified by Phage Display is capable of protecting animals against experimental infections. In the first chapter of this dissertation, we present results referring to bovine cysticercosis and show that some phages obtained can differentiate positive and negative sera. In chapter 2 we evaluate the protection of NC-1 in mice experimentally infected with T. crassiceps. Of interest, this peptide showed a protection of 74%. We conclude that Phage Display technology is capable of identifying immunoprotective peptides which were effective in experimental immunization programmes.
