Análise histológica de ovos de Macrobrachium rosenbergii (De Man) incubados em diferentes salinidades

Resumo: A espécie de camarão de água doce de maior interesse econômico atualmente é o Macrobrachium rosenbergii, um dos maiores camarões de água doce conhecido e com isso, sua produção a nível mundial é de aproximadamente 234 mil toneladas. Sua reprodução tem sido estudada com finalidades comerciais, A espécie possui características de incubar os ovos no abdômen, ficando ali aderidos até a eclosão em forma de larvas zoea. O processo de incubação dos ovos ou desenvolvimento embrionário, dura em torno de vinte dias. Neste período o embrião se desenvolve com reserva alimentar que se encontra no ovo. A vitelogenina presente no ovo está entre as proteínas mais importantes na reprodução, atuando como um dos desencadeadores da cadeia de sinalizadores endócrinos regulando a biossíntese de proteínas. Para podermos inferir a absorção de vitelogenina pelo embrião, ao longo das fases embriológicas, foram utilizadas técnicas histológicas. Para isso os ovos foram coletados de 5 em 5 dias por período de até 23 dias, separados conforme seu estádio de desenvolvimento, e processados com a técnica de histologia para visualizar o as mudanças estruturais com relação a vitelogenina utilizada pelo embrião durante seu desenvolvimento com a técnica de coloração H.E. Os ovos foram incubados em tratamentos de salinidades 0‰, 6‰ e 12‰, com temperatura média de 28ºC. A partir da obtenção de imagens das lâminas histológicas montadas com os ovos nas diferentes fases de desenvolvimento embrionário foi possível estimar a quantidade de vitelo absorvida pelo embrião a partir da observação do espaço ocupado pelo embrião no ovo em relação ao espaço previamente ocupado pelo vitelo. As imagens obtidas foram submetidas a análise com aumento de 20 vezes, posteriormente submetidos ao processador de imagens (Image pro-plus®). De acordo com os dados obtidos após tratamento das informações no referido programa e com auxílio de outro software de estatística (Statistic®). Houve diferenciação na dinâmica de absorção, pela diferenciação das estruturas nas diferentes salinidades aplicadas. Desta forma é possível inferir que o transporte acontece no interior do ovo, possibilitando o rearranjo das biomoléculas. Ainda é possível visualizar que os tratamentos diferem entre si, quando comparados ao tamanho dos espaços formados e do deslocamento do material, sendo que para os tratamentos de salinidade intermediário (6 ‰) ocorreram de forma mais rápida em relação aos outros tratamentos. Abstract: The freshwater shrimp species of major economic interest today is Macrobachium rosenbergii, one of the largest known freshwater prawns therefore, its worldwide production is approximately 234 thousand tons. Its reproduction has been studied with commercial purposes. The species has characteristics of incubating the eggs in the abdomen, being adhered there until the hatching in the form of zoea larvae. The process of egg incubation or embryo development lasts for about twenty days. In this period the embryo develops with food reserve that is in the egg. Vitelogenin present in the egg is among the most important proteins in reproduction, acting as one of the chain triggers of endocrine signals regulating protein biosynthesis. In order to infer the absorption of vitellogenin by the embryo, during the embryological phases, histological techniques were used. The eggs were collected every 5 days for a period of up to 23 days, separated according to their stage of development, and processed with the histology technique to visualize the structural changes with respect to vitellogenin used by the embryo during its development with the HE staining technique The eggs were incubated in 0 ‰, 6 ‰ and 12 ‰ salinity treatments, with a mean temperature of 28 ° C. It was possible to estimate the amount of yolk absorbed by the embryo from the observation of the space occupied by the embryo in the egg in relation to the space previously occupied by the yolk, from the images obtained from the histological slides assembled with the eggs in the different stages of embryonic development. The images were submitted to analysis with a 20-fold increase, later submitted to the image processor (Image pro-plus®). According to the data obtained after processing the information and with the help of other statistical software (Statistic®). There was differentiation in the dynamics of absorption, by the observation of the structures in the different applied salinities. Therefore, it is possible to infer that the transport happens inside the egg, making possible the rearrangement of biomolecules. It is still possible to visualize that the treatments differed from each other when compared to the size of the formed spaces and the displacement of the material, whereas for the treatments of intermediate salinity (6 ‰) they occurred faster than the other treatments.
