dc.description.abstractResumo: A família Gesneriaceae (150 gêneros / 3.500 spp) está representada no Brasil por 27 gêneros e 211 espécies. Sinningia é o gênero mais abundante no país, com 69 espécies. Várias classes de metabólitos secundários, como quinonas, terpenos, e glicosídeos fenólicos ocorrem em Sinningia spp. Visando contribuir para o conhecimento químico deste gênero, o presente trabalho descreve o estudo fitoquímico e a atividade citotóxica de S. leucotricha e S. canescens, duas espécies encontradas no estado do Paraná e conhecidas como rainha-do-abismo. Os tubérculos secos foram extraídos com solventes e os extratos fracionados através de técnicas cromatográficas como CCD, CC e CLAE. Diversos métodos espectroscópicos (UV, DC, RMN, EM, CG-EM), cálculos teóricos, e comparação com dados da literatura, foram utilizados para identificação das substâncias isoladas. No total o estudo fitoquímico forneceu 35 substâncias: 7-hidroxi- - dunniona (S1), 6-metoxi-7-hidroxi- -dunniona (S2), 7,8-dimetoxi- -dunniona (S3), 7- metoxi-8-hidroxi- -dunniona (S4), 6,7-dimetoxi- -dunniona (S5), 7,8-dimetoxidunniona (S6), 7,8-dimetoxicanescenona (S7), 7,8-dimetoxidunniol (S8), 1-hidroxi-1- metilenocarboxi-7,8-dimetoxi- -dunniona (S9), tectoquinona (S10), 2-(4'-hidroxifenil)- 5-hidroxi-7-metoxi-3-benzofuranal (S11), 2-(4'-metoxifenil)-5-hidroxi-7-metoxi-3- benzofuranal (S12), presilphiperfolan-9-ol (S13), ácido leucotrichóico (S14), leucotrichol (S15), 11-epi-subergorgiol (S16), leucotrichanal (S17), 11-episilphiperfolenal (S18), cedrol (S19), 2,6,7-tri-epi-cedrol (S20), hederagenina (S21), ácido 23-hidroxiursólico (S22), ácido betulínico (S23), halleridona (S24), cleroindicina B (S25), cleroindicina C (S26), cornosídeo (S27), 4-hidroxibenzoato de metila (S28), 4-hidroxibenzoato de etila (S29), etilglucose (S30), calceolariosídeo B (S31), calceolariosídeo A (S32), plantainosídeo A (S33), conandrosídeo (S34), e isoverbascosídeo (S35). Adicionalmente, a análise por CG-EM de frações de S. canescens levou à identificação de mais 39 sesquiterpenos, destacando-se - cedreno, 2-epi-funebreno, -cedreno e -santaleno. As substâncias S2, S3, S4, S5, S6, S7, S8, S9, S11, S12, S14, S15, S16, S17, S18 e S20 são inéditas. Os extratos etanólicos, frações e substâncias isoladas foram testados contra várias linhagens de células tumorais humanas, usando o método da sulforrodamina B. A potência foi determinada como a concentração capaz de inibir totalmente o crescimento celular (TGI). O extrato etanólico de S. canescens inibiu principalmente o crescimento das linhagens MCF-7 (mama) e NCI-ADR/RES (ovário resistente a múltiplos fármacos), com TGI de 27,82 e 28,87 g mL-1, respectivamente. Por outro lado, o extrato de S. leucotricha apresentou uma forte atividade contra as células de leucemia (K562 - TGI 0,36 g mL-1). Dentre as substâncias isoladas, S1, S14, S16, S25, S26 e S31 foram testadas. S1 foi a mais potente, inibindo o crescimento das células de melanoma (UACC-62), glioma (U251), mama (MCF-7), ovário resistente a múltiplos fármacos (NCI-ADR/RES), rim (786-0), pulmão (NCI-H460), próstata (PC-3), e leucemia (K562) com valores de TGI entre 4,58 e 31,31 g mL-1. dc.description.abstractAbstract: The Gesneriaceae family (150 genera / 3.500 spp) is represented in Brazil by 27 genera and 211 species. Sinningia is the most abundant genus in the country, with 69 species. Many classes of secondary metabolites, like quinones, terpenes, and phenolic glycosides occur in Sinningia species. Aiming to contribute for the chemical knowledge of this genus, the present work describes the phytochemical study and cytotoxic activity of S. leucotricha and S. canescens, two species found in Paraná state and known as "rainha-do-abismo". Dried tubers were extracted with solvents and the extracts were fractionated by chromatographic techniques, like PTLC, CC and HPLC. Several spectroscopic methods (UV, CD, NMR, MS, GC-MS), theoretical calculations, and comparison with the literature, were used to identify the isolated substances. This phytochemical study resulted in 35 compounds: 7-hydroxy- - dunnione (S1), 6-methoxy-7-hydroxy- -dunnione (S2), 7,8-dimethoxy- -dunnione (S3), 7-methoxy-8-hydroxy- -dunnione (S4), 6,7-dimethoxy- -dunnione (S5), 7,8- dimethoxy-dunnione (S6), 7,8-dimethoxy-canescenone (S7), 7,8-dimethoxy-dunniol (S8), 1-hydroxy-1-methylenecarboxy-7,8-dimethoxy- -dunnione (S9), tectoquinone (S10), 2-(4'-hydroxyphenyl)-5-hydroxy-7-methoxy-3-benzofuranal (S11), 2-(4'- methoxyphenyl)-5-hydroxy-7-methoxy-3-benzofuranal (S12), presilphiperfolan-9-ol (S13), leucotrichoic acid (S14), leucotrichol (S15), 11-epi-subergorgiol (S16), leucotrichanal (S17), 11-epi-silphiperfolenal (S18), cedrol (S19), 2,6,7-tri-epi-cedrol (S20), hederagenin (S21), 23-hydroxyursolic acid (S22), betulinic acid (S23), halleridone (S24), cleroindicin B (S25), cleroindicin C (S26), cornoside (S27), methyl 4-hydroxybenzoate (S28), ethyl 4-hydroxybenzoate (S29), ethylglucoside (S30), calceolarioside B (S31), calceolarioside A (S32), plantainoside A (S33), conandroside (S34), and isoverbascoside (S35). Additionally, GC-MS analysis of the fractions from S. canescens, lead to identification of more 39 sesquiterpenes, highlighting: -cedrene, 2-epi-funebrene, -cedrene and -santalene. Compounds S2, S3, S4, S5, S6, S7, S8, S9, S11, S12, S14, S15, S16, S17, S18 and S20 are new. Ethanolic extracts, fractions and isolated substances were tested against several human tumor cell lines using the sulphorrodamin B method. The level of activity of the extracts was determined as the concentration that caused total growth inhibition of the cells (TGI). S. canescens ethanolic extract inhibited mainly the growth of MCF-7 (breast) and NCI-ADR/RES (ovary resistant to multiple drugs) cell lines, with TGI values of 27.82 and 28.87 g mL-1, respectively. On the other hand, S. leucotricha extract showed a strong activity against leukemia (K562 - TGI 0.36 g mL-1) cell lines. Among isolated compounds, S1, S14, S16, S25, S26 and S31 were tested. S1 was the most active, inhibiting the growth of melanoma (UACC-62), glioma (U251), breast (MCF-7), ovary resistant to multiple drugs (NCI-ADR/RES), kidney (786-0), lung (NCI-H460), prostate (PC-3), and leukemia (K562) cell lines, with TGI values varying from 4.58 to 31.31 g mL-1.
