Biotransformaçao por fungos do ácido betulínico e derivados

Resumo: Modificações nas estruturas de moléculas complexas podem ser realizadas através de sistemas envolvendo microrganismos. O ácido betulínico (I), o ácido betulônico (II) e a betulina (III), por possuírem essa característica e apresentarem importantes atividades farmacológicas, foram escolhidos para serem biotransformados por fungos. Com a finalidade de se obter os substratos, fez-se o estudo fitoquímico da casca do caule de Platanus orientalis L., com o isolamento e a identificação de três substâncias: ácido betulínico (I); ácido diidrobetulinico (XIV) e ?-sitosterol (XIII). O ácido diidrobetulínico é inédito como composto natural. O ácido betulônico foi obtido por derivatização do ácido betulínico. Para obter os fungos necessários para a biotransformação, foram isolados e identificados os fungos epifíticos e endofíticos associados à casca de P. orientalis. Após realização de experimentos preliminares foram selecionados os fungos Arthobotrys sp., Chaetophoma sp., Colletotrichum sp. e Dematium sp. A biotransformação do ácido betulínico pelo fungo Chaetophoma sp. e Dematium sp. resultou na sua oxidação levando ao ácido betulônico (II); pelo Colletotrichum sp. resultou no ácido 3-oxo,15?-hidroxilup-20(29)-en-28-óico. O ácido betulônico (II) foi biotransformado pelos fungos: Arthobotrys sp. gerando os metabólitos ácido 3-oxo, 7?-hidroxilup-20(29)-en-28-óico, ácido 3-oxo, 7?,15?- diidroxilup-20(29)-en-28-óico e ácido 3-oxo, 7?,30-diidroxilup-20(29)-en-28-óico; Chaetophoma sp. formando o ácido 3-oxo, 12?-hidroxilup-20(29)-en-28-óico, ácido 3-oxo, 7?,25-diidroxilup-20(29)-en-28-óico e Colletotrichum sp. produzindo o ácido 3- oxo, 15?-hidroxilup-20(29)-en-28-óico e o ácido 3-oxo, 7?,15?-diidroxilup-20(29)-en- 28-óico. A betulina (III) foi biotransformada pelo fungo Chaetophoma sp. que levou a formação da substância 3-oxo-lup-20(29)-en-28-ol. Portanto, através da biotransformação dos substratos propostos, foram obtidos 6 metabólitos inéditos, além do ácido betulônico e do ácido 3-oxo,7?,15?- diidroxilup-20(29)-en-28-óico. Abstract: Complex molecules can be modified through microbial transformation. The betulinic acid (I), betulonic acid (II) and betulin (III), for this characteristic and their important pharmacologic activities, have been chosen to be biotransformed by fungi. In order to obtain the compounds, the phytochemical study of Platanus orientalis L. bark was done and allowed the isolation and identification of three components: betulinic acid (I); dihydrobetulinic acid (XIV) and ?-sitosterol (XIII).The dihydrobetulinic acid is unknown as a natural compound. To get the necessary fungi for the biotransformation, it was isolated and identified epiphytes and endophytes fungi associated with the P.orientalis bark. After accomplishment of screening the Arthobotrys sp., Chaetophoma sp., Colletotrichum sp. and Dematium fungi were selected. The biotransformation of the betulinic acid by Chaetophoma sp. and Dematium sp. fungi through its oxidation resulted in betulonic acid (II) and for the Colletotrichum sp. resulted in 3-oxo-15?-hydroxy-lup-20(29)-en-28-oic acid. The betulonic acid (II) was biotransformed by the fungi : Arthobotrys sp. resulted in metabolites which were identified as 3-oxo-7?-hydroxy-lup-20(29)-en-28-oic acid, 3- oxo-7?,15?-dihydroxy-lup-20(29)-en-28-oic acid and 3-oxo-7?,30-dihydroxy-lup- 20(29)-en-28-oic acid; Chaetophoma sp. resulted in 3-oxo-12?-hydroxy-lup-20(29)- en-28-oic acid and 3-oxo-7?,25-dihydroxy-lup-20(29)-en-28-oic acid; and Colletotrichum sp. resulting in 3-oxo-15?-hydroxy-lup-20(29)-en-28-oic acid and 3- oxo-7?,15?-dihydroxy-lup-20(29)-en-28-oic acid. The betulin (III) was biotransformed by the fungi Chaetophoma sp. resulted in 3-oxo-lup-20(29)-en-28-ol. Summarising, through the biotransformation of proposed compounds six unknown metabolites, beyond the betulonic acid and 3-oxo, 7?, 15?-dihydroxy-lup- 20(29)-en-28-oic acid were identified.
