dc.description.abstractResumo: A família de pequenas GTPases monoméricas compreende um grupo de moléculas que têm sua atividade regulada pela transição entre uma forma ativa ligada a GTP e uma forma inativa ligada a GDP. A subfamília de GTPases Rnd (Rnd1, Rnd2 e Rnd3) foram caracterizadas como um grupo diferente das outras Rho GTPases, pois possuem pouca atividade GTPásica intrínseca e encontram-se constitutivamente ativas. As proteínas Rnd são expressas em diferentes tecidos: Rnd1 foi identificada em fígado, cérebro e em miométrio humano durante a gravidez. Rnd2 é expressa em testículos e Rnd3 é ubiquamente expressa. Rnd1 está envolvida em uma variedade de mecanismos moleculares relacionados com a plasticidade do citoesqueleto de actina e dinâmica dos microtúbulos. A atividade de Rnd1 está envolvida com a extensão inicial de processos neuríticos em células PC-12 e com o alongamento de espinhos dendríticos e desenvolvimento de dendritos em neurônios de hipocampo de ratos. Compreender os mecanismos que regulam a atividade de Rnd1 é um desafio. A proteína STI1 foi descrita como uma proteína envolvida na formação de pseudopodia em associação com o citoesqueleto de actina, chamando por esta razão a nossa atenção. Entretanto, STI1 foi melhor caracterizada como uma molécula relacionada ao estresse térmico em leveduras e então denominada Stress-Inducible Protein 1 (STI1) e seu homólogo em humano Hop (Heat-shock organizing protein). Esta proteína desencadeia sinais intracelulares neuroprotetores e promove a neuritogênese em células neurais por interagir com a proteína prion celular. Assim, para investigar nossa hipótese ensaios de pulldown com diversas GTPases recombinantes, co-imunoprecipitação e ensaios de interação em placas de poliestireno foram realizados para avaliar a especificidade da interação entre Rnd1 e STI1. Células PC-12, N2a e SHSY-5Y foram utilizadas para ensaios de neuritogênese e diferenciação celular, enquanto células COS-7 foram empregadas em ensaios de contração celular. Lipid rafts foram purificados a partir de encéfalos de camundongos e células PC-12 biotiniladas na superfície celular, com o propósito de analisar a distribuição das proteínas Rnd1 e STI1. Os ensaios bioquímicos sugerem que Rnd1 e STI1 interagem in vitro. A expressão da proteína GFP-STI1 reverte a neuritogênese desencadeada pela proteína GFP-Rnd1 em células PC-12, N2a e SHSY-5Y e induz diferenciação em PC-12. A expressão da quimera GFP-STI1 e a proteína recombinante 6His- STI1 reduziram a contração celular em células COS-7 mediada pela interação entre GFP-Rnd1 e a porção citoplasmática da plexina-A1. STI1 e Rnd1 estão localizadas tanto nos lipid rafts quanto nas frações solúveis de membrana de encéfalos de camundongos. Entretanto, STI1 encontra-se predominantemente na porção intracelular dos lipid rafts de células PC-12. Os resultados demonstram que as proteínas Rnd1 e STI1 interagem e que tal interação promove a regulação da plasticidade do citoesqueleto.pt_BR
dc.description.abstractAbstract: The small GTPase family of proteins comprises a group of molecules that have their activity regulated by switching between an active form GTP bound and inactive when hydrolyses the GTP to GDP. The Rnd family (Rnd1, Rnd2 and Rnd3) was first characterized as a different branch of the Rho GTPase family that has low affinity for GDP and are in a constitutively activated form. The Rnd proteins are expressed in different tissues: Rnd1 was found in liver, brain and human myometria during pregnancy. Rnd2 is expressed in testis and Rnd3 has a ubiquitously low level expression and are mainly involved in a variety of molecular mechanisms related to rearrangements of the actin cytoskeleton and microtubule dynamics. Rnd1 activity has been involved with process extension in PC-12 cells and with the elongation of dendritic spines and dendrite development in rat hippocampal neurons.Understand the mechanisms that regulate its activity are a challenge. STI1 was first characterized as a molecule related to yeast stress and so called yeast-stress-inducible protein 1 (STI-1) and its homologous proteins were then identified as the co-chaperone murine Stress-Inducible protein 1 (mSTI1) and Hop (Heat-shock organizing protein) in human. This protein triggers neuroprotective signals and promotes neuritogenesis by its characterized interaction with the prion protein. STI1 was also characterized as a molecule that was associated with pseudopodia formation in association with actin cytoskeleton. So, the main purpose of this work was biochemically characterize the interaction between Rnd1 and STI1 and analyze its biological function in the regulation of cytoskeleton plasticity and neuronal modulation. Then, to investigate our hypothesis pulldown, coimmunoprecipitation and binding assays were performed to evaluate the specificity of the interaction between Rnd1 and STI1. PC-12, N2a and SHSY-5Y cells were used for neuritogenesis assays and differentiation assays, while COS-7 cells were employed for cell contraction analysis. Lipid rafts were purified from mice brain and from surface biotinylated PC-12 cells for analysis of STI1 and Rnd1 distribution. Biochemical assays suggested that Rnd1 and STI1 can interact in vitro. Cellular GFP-STI1 expression reverts the neuritogenesis induced by GFP-Rnd1 on PC-12, N2a and SHSY-5Y cells and induces PC-12 differentiation. GFP-STI1 expression or soluble recombinant 6His-STI1 decreases COS-7 cell contraction mediated by GFP-Rnd1 and cytoplasmic Plexin-A1. STI1 and Rnd1 are present both in lipid rafts and detergent soluble membrane fractions from mouse brain. PC-12 biotinylated rafts indicated that STI1 could be present in the cytoplasmic side of rafts. Results show that Rnd1 and STI1 interact to each other and this interaction is involved with cytoskeleton plasticity.pt_BR
