Resolução enzimática de alcoóis secundários

Resumo: Neste trabalho foi testada a capacidade de resolução de três lipases diferentes em álcoois secundários. As lipases estudadas foram de, Rhizomucor miehei (Lipozyme RM IM, Novozymes) Thermomyces lanuginosus (Lipozyme TL IM, Novozymes) e Burkholderia cepacia, sendo as duas primeiras comerciais e a última produzida no Laboratório de Tecnologia Enzimática e Biocatálise do Departamento de Química da UFPR. Este estudo foi realizado empregando-se a enzima de Candida antarctica (Lipozyme CAL-B IM, Novozymes), como referência. A enzima que obteve menor desempenho em termos de velocidade de reação e enantiosseletividade foi à de Rhizomucor miehei e a de maior desempenho nos mesmos parâmetros foi a de Candida antarctica.
Outro estudo realizado neste trabalho foi a resolução de álcoois secundários envolvendo a esterificação química da mistura racêmica dos álcoois com posterior hidrólise enzimática utilizando-se as enzimas de Thermomyces lanuginosus e Candida antarctica. Os melhores resultados foram obtidos com a enzima de Candida antarctica.
Devido a dificuldades na obtenção de tais enzimas, resolveu-se estudar a resolução de álcoois alílicos terminais com Candida antarctica. Os testes de resolução enzimática apresentaram boa estereosseletividade, porém ressaltou-se uma melhor eficiência em termos de velocidade de reação e enantiosseletividade para as moléculas aromáticas. Tais resultados abrem perspectivas para um trabalho envolvendo a síntese de -iodo epóxidos quirais, importantes intermediários de reações na síntese de feromônios de lepidópteros. Abstract: In this work the capacity of resolution of three lipase different in secondary alcohols was tested. Lipase studied were of, Rhizomucor miehei (Lipozyme RM IM, Novozymes) Thermomyces lanuginosus (Lipozyme TL IM, Novozymes) and Burkholderia cepacia, being the two first ones commercial and the last one produced in the Laboratório de Tecnologia Enzimática e Biocatálise - Department of Chemistry - UFPR. This study was carried through using the enzyme of Candida antarctica (Lipozyme CAL-B IM, Novozymes), as reference. The enzyme that lead to the poorest performance in terms of rate reaction and enantioselectivity was the one of Rhizomucor miehei and the one of the best performance in the same parameters was the Candida antarctica.
Another study carried through in this work was the resolution of secondary alcohols involving the chemical sterification of the racemic mixture of the alcohols with posterior enzymatic hydrolysis using enzymes of Thermomyces lanuginosus and Candida antarctica. The best results were obtained with the enzyme of Candida antarctica.
Due to the difficulties in the attainment of such enzymes, it was decided to study the resolution of terminal allyl alcohols with Candida antarctica. The tests of enzymatic resolution presented good stereoselectivity, however a better efficiency in terms of rate reaction and enantioselectivity for aromatic molecules standed out. Such results open out perspectives for a work involving the synthesis chirals -iodo epoxides, an important intermediate in the synthesis of lepdoptera pheromones.
