Estabilidade físico-química da fase aquosa resultante do fracionamento do bio-óleo

Resumo: O bio-óleo é uma fonte promissora para geração de produtos de maior valor agregado. Uma das formas de simplificar o aproveitamento das diversas potencialidades do bio-óleo é o fracionamento, que consiste na separação de fases que o constituem. A fase insolúvel, denominada lignina pirolítica tem aplicações conhecidas pela literatura, enquanto a fase solúvel resultante carece de informações acerca do comportamento físico-químico, atividade antioxidante e antimicrobiana. Desse modo, o presente estudo objetivou caracterizar quimicamente a fase aquosa (FA) resultante do fracionamento do bio-óleo e verificar sua estabilidade físico-química, bem como seu potencial antioxidante e antimicrobiano. Para tal, o bio-óleo foi fracionado em água fria a 17.000 RPM em três proporções bio-óleo: água, sendo elas 1:100, 1:70 e 1:50. A fase aquosa resultante do fracionamento foi caracterizada em relação as propriedades físico-químicas, composição química via cromatografia gasosa acoplada a espectrometria de massas, estabilidade físico-química sob condições de envelhecimento, além da potencial capacidade antioxidante e ação biocida contra fungos e bactérias. Os principais resultados do presente estudo mostraram que quanto menor o teor de água utilizado no fracionamento, maior é o rendimento, o qual foi superior (em torno de 60%) para a fração 1:50. Observou-se a presença de classes de compostos químicos na fase aquosa, sendo eles éteres, ésteres, aldeídos, compostos nitrogenados, anidro açucares, fenóis, álcoois e alcanos, com destaque para o catecol, o 5-hidroximetilfurfural, a vanilina e a levoglicosana. Especificamente, a levoglicosana foi o composto majoritário, atingindo cerca de 44% em área relativa na proporção 1:50. A fase aquosa apresentou relevante capacidade antioxidante em decorrência da sua composição química fortemente fenólica, que somada a outros compostos químicos detectados na fase aquosa resultaram também em uma alta atividade antifúngica contra os fungos Trametes versicolor e Gloeophyllum trabeum, e contra as bactérias gram-negativa Escherichia coli e gram-positiva Staphylococcus aureus. De maneira geral, os compostos químicos detectados na fase aquosa se mostraram estáveis quimicamente ao longo de 43 dias de envelhecimento, confirmando que a fase aquosa pode ser considerado uma promissora alternativa para a extração e purificação de químicos de maior valor agregado em um sistema de biorrefinaria, bem como aplicações como agente antimicrobiano e antioxidante. Abstract: Bio-oil is a promising source for the generation of higher value-added products. One of the ways to simplify the use of the various potentialities of bio-oil is the fractionation, which consists of the separation of phases through methods such as liquid-liquid extraction, molecular distillation and extraction in cold water, the latter being the most advantageous due to the simplicity of processing. The insoluble phase, called pyrolytic lignin has applications known in the literature, while the resulting water-soluble phase lacks information about physicochemical behavior, antioxidant and antimicrobial activity. Thus, the present study aimed to chemically characterize the FA and verify its physicochemical stability, as well as its antioxidant and antimicrobial potential. For this, the bio-oil was fractionated in cold water at 17,000 RPM in three bio-oil: water ratios, being them 1:100, 1:70 and 1:50. The aqueous phase resulting from the fractionation was characterized in relation to physicochemical properties, chemical composition via gas chromatography coupled to mass spectrometry, physicochemical stability under aging conditions, the potential antioxidant capacity and biocide action against fungi and bacteria. The main results of this study showed that the lower the water content used in the fractioning, the higher the yield, which was higher (around 60%) for the 1:50 fraction. It was observed the presence of chemical compounds in the water-soluble phase, being them ethers, esters, aldehydes, nitrogen compounds, anhydrous sugars, phenols, alcohols and alkanes, with emphasis on catechol, 5-hydroxymethylfurfural, vanillin and levoglucosan. Specifically, levoglucosan was the major compound, reaching about 44% in relative area in the proportion 1:50. The water-soluble phase showed relevant antioxidant capacity, especially due to its chemical phenolic composition. These phenolic compounds, as well as other chemical compounds detected in the water-soluble phase resulted in a high antifungal activity against the fungi Trametes versicolor and Gloeophyllum trabeum, and against the gram-negative bacteria Escherichia coli and gram-positive Staphylococcus aureus. In general, the chemical compounds detected in the water-soluble phase have shown to be chemically stable over 43 days of ageing, confirming that the water-soluble phase can be considered a promising alternative for the extraction and purification of higher added value chemicals in a biorefinery system, as well as applications as an antimicrobial and antioxidant agent.
