Participação de receptores NK1 nas respostas inflamatória e febril induzidas por endotoxina bacteriana
Abstract
Resumo: As respostas inflamatória e febril são importantes para a defesa do organismo contra agentes agressores. Mediadores inflamatórios liberados por células do sistema imune e neurotransmissores e peptídeos provenientes de fibras nervosas, especialmente das fibras vagais, são importantes na interligação destas respostas. Estudos anteriores realizados em nosso laboratório evidenciaram a importância do nervo vago e da acetilcolina, o principal neurotransmissor das fibras eferentes vagais, na manutenção das células residentes na cavidade peritonial e, então, na modulação das respostas inflamatória e febril. A substância P (SP) é um neuropeptídeo abundante nas fibras aferentes vagais e dados da literatura sugerem que a mesma é capaz de modular a liberação de citocinas, bem como a liberação de outros mediadores inflamatórios pelas células do sistema imune, ao se ligar aos receptores para taquicininas NK1. No entanto, faltam estudos in vivo que relacionem as conseqüências desses efeitos mediados pela substância P nas respostas inflamatória e febril. Neste trabalho avaliou-se o papel deste neuropeptídeo nestas respostas, testando-se o efeito do antagonista seletivo de receptor NK1, SR140333, nas respostas inflamatórias (extravasamento plasmático e migração celular) e febril, induzidas por lipopolissacarídeo (LPS). Os resultados mostraram que a administração sistêmica de SR140333 não é capaz de modificar o número de células residentes na cavidade peritonial; a febre, o extravasamento plasmático ou a migração celular induzidos por LPS, sugerindo que a SP liberada perifericamente não é essencial para o desencadeamento dessas respostas induzidas por esta endotoxina. No entanto, a administração central de SR140333 reduziu significativamente a febre induzida pela administração periférica de LPS. Esses dados evidenciam um papel relevante da SP presente no sistema nervoso central na resposta febril desencadeada pelo LPS. Em adição, a administração intracerebroventricular de substância P, em animais tratados com captopril, induziu uma resposta febril que foi significativamente reduzida pelo SR140333. Considerando que muitos mediadores estão envolvidos na resposta febril induzida por LPS, como a interleucina (IL)-1ß, avaliou-se o efeito do SR140333 na febre induzida por esta citocina, que é considerada um dos principais pirogênios endógenos. Porém, diferente do que foi observado para o LPS, o SR140333 não modificou a febre induzida pela IL-1ß. Desta forma, é possível que a SP esteja sendo liberada antes da IL-1ß na cascata de mediadores da febre do LPS ou, ainda, que a SP esteja atuando na via de outros mediadores envolvidos na resposta febril desta endotoxina. Abstract: The inflammatory and febrile responses are essential components of host defense against environmental aggressors such as pathogens. Inflammatory mediators released by the immune cells, along with peptides and neurotransmitters, mainly from the vagus nerve, play a key role in these neuro-immune interactions. Previous studies in our lab have shown an important role of the acetylcholine released from the efferent fibers of the vagus nerve on the maintenance of peritoneal resident cells and consequently in the modulation of inflammatory and febrile responses. Substance P (SP) is an abundant neuropeptide of the afferent vagus fibers, and data from literature suggest that SP is able to modulate apoptosis and inflammatory mediators release such as cytokines and others, through activation of tachykinins NK1 receptors. However, there is a lack of in vivo studies evaluating the role of SP in inflammatory and febrile responses. In this study we investigated the effect of the selective NK1 receptor antagonist, SR140333, on the inflammatory (protein extravasation and cell migration) and febrile responses induced by lipopolyssacharide (LPS). The results showed that the systemic injection of SR140333 did not modify the fever, protein extravasation, or the cell migration to the peritoneal cavity induced by LPS, suggesting that peripheral SP is not essential to the development of these responses. However, intracerebroventricular administration of SR140333 significantly reduced the fever induced by peripheral injection of LPS. These data emphasize an important role of the SP on the central nervous system during the febrile response induced by this endotoxin. In addition, intracerebroventricular injection of SP in captopril-treated rats induced a febrile response that was significantly reduced by SR140333. Considering that there are many mediators involved in the LPS-induced fever, such as interleukin (IL)-1ß, we also evaluated the effect of SR140333 on the fever induced by this cytokine which is considered one of the most important endogenous pyrogens. Surprisingly, different from what we observed after LPS, SR140333 did not modify the fever induced by IL1ß. Therefore, it is possible that SP is released previously to IL-1ß, or that SP acts on different pathways involved in LPS-induced fever. Altogether these data suggest that, different from what we have observed for acetylcholine, the substance P releasing peripherally is not relevant for fever, neutrophil migration or protein extravasation induced by LPS. However, central release of substance P is essential for LPS-induced fever, but not for IL-1ß.
