Cicloadição 1,3-dipolar interrompida catalisada por Cu(I) na síntese de 5-organocalcogeno (S/Se/Te)-1,2,3-triazóis usando acetiletos de Ag(I)

Resumo: A presente tese se refere ao desenvolvimento de uma nova metodologia para a síntese de 5-organocalcogeno-1,2,3-triazol-1,4,5-trissubstituídos. Essa metodologia consiste na reação de cicloadição 1,3-dipolar ininterrupta catalisada por Cu(I) entre azidas orgânicas e acetiletos de Ag(I) na presença de dicalcogenetos de diorganoila (S/Se/Te). A melhor condição de reação para a obtenção dos produtos desejados com rendimentos entre 15-92 % envolveu o uso de 0,55 mmol de azidas orgânicas, 0,250 mmol dos dicalcogenetos de diorganoila, na presença de CuI (10 mol %) e 1,10-fenantrolina (1,10-phen) (20 mol %), em sulfóxido de dimetila (DMSO) como solvente, cloreto de lítio (LiCl) (2,2 equiv.) como aditivo e 0,50 mmol dos correspondentes acetiletos de Ag(I), adicionados em 3 porções (0,50 mmol/3 = 0,166 mmol a cada 30 minutos), sob atmosfera de argônio e a temperatura ambiente. A metodologia sintética descrita nessa tese foi efetiva para inserção de fragmentos orgânicos de S/Se/Te na posição 5 do núcleo 1,2,3-triazol, mas limitada para a selanilação utilizando disseleneto de di(2,4,6-trimetil)fenila e disseleneto de di(1 -naftila), fornecendo os produtos em rendimentos baixos. No que diz respeito a sulfanilação da posição 5 do núcleo triazólico utilizando dissulfeto de difenila sob a condição de reação otimizada, baixos rendimentos foram obtidos. Por exemplo, para alcançar moderados 54% de rendimento, foi necessário a utilização de 50 mol % de CuI e 100 mol % de 1,10-phen. Entre os 34 compostos obtidos, 20 são inéditos, o que demonstra a versatilidade e robustez desse método. Embora o uso de acetiletos de Ag(I) tenham sido menos explorados devido baixa reatividade/e ou solubilidade, o uso deles na reação de cicloadição 1,3-dipolar interrompida catalisada por Cu(I) foi promissor para a obtenção de 5-organocalcogeno-1,2,3- triazóis-1,4-dissubstituídos. Quanto a solubilidade, a adição de LiCl em uma solução de acetileto de Ag(I) em DMSO é essencial para romper as interações argentofílicas da rede polimérica e promover a solubilidade total sob as condições de reações. Embora os experimentos qualitativos relatados nesse trabalho sejam incipientes, o sistema LiCl/DMSO tem um efeito sinérgico no aumento da solubilidade do acetileto de Ag(I) e possibilitam o desenvolvimento de novas metodologias sintéticas utilizando diretamente esses reagentes organometálicos. Abstract: The present thesis is related to the development of a new methodology for the synthesis of 5-organochalcogeno-1,2,3-triazol-1,4,5-trisubstituted using silver acetylides (RCCAg), organic azides, and diorganoyl dichalcogenides (S/Se/Te) as starting reagents. This methodology involves the Cu(I)-catalyzed interrupted azide-silver acetylides 1,3-dipolar cycloaddition reaction (Interrupted CuAAC) in the presence of diorganoyl dichalcogenides (S/Se/Te) as electrophilic species. The best reaction condition to obtain the desired products with yields ranging from 15-92 % involved the use of 0.55 mmol of organic azides, 0.250 mmol of diorganoyl dichalcogenides, in the presence of Cul (10 mol %) and 1,10- phenanthroline (1,10-phen)(20 mol %), in dimethyl sulfoxide (DMSO) as the solvent, lithium chloride (LiCl) (2.2 equiv.) as additive, and 0.50 mmol of the corresponding Ag(l) acetylides added in 3 portions (0.50 mmol/3 = 0.166 mmol every 30 minutes), under argon atmosphere and at room temperature for 3 hours. The synthetic methodology described in this thesis was effective for the insertion of S/Se/Te organic fragments at 5-position of the 1,2,3-triazole scaffold but limited for selanylation using bis(2,4,6-trimethyl)phenyl diselenide and bis(1-naphthyl) diselenide, providing the products in low yields. Regarding the sulfanylation at 5- position of the triazole core using diphenyl disulfide under the optimized reaction condition, low yields were obtained. For example, to achieve a moderate 54 % yield, it was necessary to use 50 mol % of Cul and 100 mol % of phenanthroline. Among the 34 synthesized compounds, 20 are novel, demonstrating the versatility and robustness of this method. Although the use of Ag(l) acetylides has been less explored due to either low reactivity or solubility, their use in Cu(I)- catalyzed 1,3-dipolar cycloaddition reactions with organic azides has shown promise for afford 1,2,3-triazole-1,4,5-trisubstituted. Additionally, issues related to the limited solubility of Ag(l) acetylides were solved. The addition of LiCl to a solution of Ag(l) acetylide in DMSO promoted the complete solubility of (RC^CAg)n. Although the qualitative experiments reported in this work suggest that the DMSO/LiCl system has a synergistic effect on increasing the solubility of Ag(l) acetylide, the described results enable the development of new methodologies using Ag(l) acetylides.
