Identificação de terpenóides como feromônios produzidos por percevejos das espécies Orsilochides leucoptera (Hemiptera: Scutelleridae) e Mormidea V-Luteum (Hemiptera : Pentatomidae)

Resumo: Neste trabalho é descrito o estudo de aspectos relacionados à ecologia química de duas espécies de percevejos, abordando a identificação da estrutura e estereoquímica dos compostos macho-específicos, assim como o estudo da bioatividade em laboratório de tais compostos por meio de testes de bioensaio. Assim, a tese apresentada está organizada em dois capítulos. No primeiro são mostrados os resultados gerados a partir do estudo da espécie Orsilochides leucoptera, cujos machos produzem os terpenoides (R)-cânfora e (R)-borneol. Tais compostos mostraram alta atratividade para as fêmeas da espécie, indicando tratar-se de um feromônio sexual. Nas glândulas metatorácicas (MTG) de machos e fêmeas foram identificados 7 compostos já descritos para outras espécies de percevejos, além de (R)-a-terpineol detectado apenas na MTG de machos, sugerindo que este composto pode ter relação à biossíntese da cânfora e borneol. O segundo capítulo trata dos estudos realizados com Mormidea v-luteum. Nesta espécie foram identificados 6 compostos macho-específicos, os quais tiveram suas estruturas propostas a partir dos seus espectros de massas e infravermelho. As estruturas sugeridas para tais compostos foram zingiberenol (2 isômeros), murgantiol (3 isômeros) e sesquipiperitol (1 isômero), sendo todas confirmadas por meio da comparação do perfil cromatográfico dos voláteis naturais e dos produtos da síntese dos respectivos racematos. A determinação da estereoquímica dos isômeros do zingiberenol e murgantiol foi realizada por meio de diferentes estratégias, sendo identificadas como (1S,4R,1’S)-zingiberenol, (1R,4R,1’S)-zingiberenol, (1R,4R,1’S,4’R)-murgantiol, (1R,4R,1’S,4’S)-murgantiol e (1S,4R,1’S,4’S)-murgantiol. O sesquipiperitol detectado provavelmente é um intermediário biossintético para os outros compostos macho-específicos. Testes de bioensaio com os isômeros de zingiberenol e murgantiol mostraram que estes agem possivelmente como feromônio sexual-agregação, sendo o primeiro relato de todos estes compostos juntos em machos de uma mesma espécie. Abstract: This project describes the study of aspects related to the chemical ecology of two species of bed bugs, approaching the identification of the structure and stereochemistry of malespecific compounds, as well as the study of the bioactivity in the laboratory of such compounds through bioassay tests. Thus, the thesis presented is organized in two chapters. The first chapter shows the results generated from the study of the species Orsilochides leucoptera, whose males produce the terpenoids (R)-camphor and (R)-borneol. These compounds showed high attractiveness for females of the species, indicating that it is a sex pheromone. In the metathoracic glands (MTG) of males and females, 7 compounds already described for other species of bugs were identified, in addition to (R)-a-terpineol detected only in the MTG of males, suggesting that this compound may be related to camphor and borneol biosynthesis. The second chapter deals with studies carried out with Mormidea v-luteum. In this species, 6 malespecific compounds were identified, which had their structures proposed from their mass and infrared spectra. The suggested structures for such compounds were zingiberenol (2 isomers), murgantiol (3 isomers) and sesquipiperitol (1 isomer), all of which were confirmed by comparing the chromatographic profile of the natural volatiles and the synthesis products of the respective racemates. The determination of the isomers stereochemistry of zingiberenol and mugantiol was performed using different strategies, being identified as (1S,4R,1'S)- zingiberenol, (1R,4R,1'S)-zingiberenol, (1R,4R,1'S,4'R)-murgantiol, (1R,4R,1'S,4'S)- murgantiol and (1S,4R,1'S,4'S)-murgantiol. Sesquipiperitol detected is probably a biosynthetic intermediate for the other male-specific compounds. Bioassay tests with the isomers of zingiberenol and mungantiol showed that they possibly act as a sex-aggregation pheromone, being the first report of all these compounds together in males of the same species.
