Caracterização de leveduras vínicas provenientes do Município de Pinto Bandeira(RS) e da Região Metropolitana de Curitiba(PR)

Resumo: O uso de leveduras Saccharomyces cerevisiae selecionadas facilita o controle do processo fermentativo, reduzindo diferenças na qualidade do vinho de uma safra para a outra. Entre as características básicas de uma linhagem selecionada estão a alta taxa fermentativa, produção baixa ou nula de H2S e neutralidade quanto ao fator killer. Leveduras autóctones, isoladas da própria região de elaboração do vinho, são preferidas por estarem melhores adaptadas às uvas da região e conferirem tipicidade ao produto. No Brasil, o vinho é elaborado em diversas regiões, como a Serra Gaúcha, no RS, principal polo vitivinícola, Vale do São Francisco, Pernambuco/Bahia, e a região Metropolitana de Curitiba, no PR. Esta última é caracterizada pela elaboração artesanal de vinho. Nestas regiões, há crescente demanda por linhagens selecionadas autóctones. Nesse contexto, o objetivo do presente trabalho foi avaliar o perfil enológico de 175 leveduras isoladas da região metropolita de Curitiba, no PR, nos anos de 2003 e 2012, e do município de Pinto Bandeira, no RS, quanto aos aspectos enológicos favoráveis à elaboração de vinho. Paralelamente, foram feitas a validação do meio Lorena/ELNC (80:20) quanto à detecção do fator killer e tentativas de melhoria de tecnologia para isolamento e detecção dos plasmídeos killer, responsável pelo fenótipo K+R+ em leveduras vínicas. A produção e detecção do fator killer e as características sensível e neutra foram avaliadas, em triplicata, em meio sólido Lorena/ELNC (80:20). A velocidade fermentativa e a produção de H2S foram avaliadas, em triplicata, em meio mosto sulfito. As linhagens com perfil para a elaboração de vinhos e fenótipo killer foram identificadas por amplificação da região ITS1- 5.8S-ITS2 por PCR. Os resultados mostraram que a maioria das linhagens apresentou baixa velocidade fermentativa, baixa ou nula produção de H2S e fenótipo K-R+. De um total de 120 leveduras isoladas,foram obtidas 16 linhagens de Saccharomyces cerevisiae com perfil enológico para o município de Pinto Bandeira. Nenhuma linhagem, dentre as 42 isoladas na região metropolitana de Curitiba, em 2012, mostrou aptidão para a elaboração de vinho. Apenas uma levedura isolada em 2003 apresentou perfil enológico adequado. Uma linhagem de cada região foi escolhida para vinificar mosto Pinot Noir, para avaliar as características físico-químicas dos vinhos elaborados. Foram analisados o teor alcoólico, açúcares redutores totais residuais, acidez volátil e pH. Em cinco dias os vinhos já se encontravam secos, com menos de 3 g.L-1 de ART, restando apenas a realização da fermentação malolática. A análise de compostos voláteis e minerais revelou, respectivamente, acentuada concentração produção de alguns álcoois superiores e alto teor de Zn. Dessa forma, as leveduras autóctones aqui isoladas e selecionadas devem conferir vantagem sobre a fermentação espontânea tradicional por elaborarem vinhos de qualidade, com a certeza de que metabólitos secundários indesejados não serão produzidos. A metodologia para o isolamento do plasmídeo responsável pela produção do fator killer foi modificada, porém novos estudos devem ser feitos para reduzir o tempo de execução. Abstract: The use of selected Saccharomyces cerevisiae yeasts allows the control of the fermentation process, reducing the differences in the wine quality from one harvest to another. Among the basic characteristics of selected strain are the high fermentation rate, low or absent production of H2S and neutrality regarding the killer factor. Autochthonous yeasts, isolated from the own winemaking region, are preferred for being better adapted to the grapes of the region and confer typicity to the product. In Brazil, the wine is elaborated in several regions, such as the Serra Gaucha, which is the main wine pole, Vale do São Francisco, PE/BA, and the Metropolitan region of Curitiba, PR. This one is characterized by artisan winemaking. In these regions, there is growing demand for autochthonous selected strains. In this context, the goal of the present work was to evaluate the enological profile of 175 yeasts isolated from metropolitan region of Curitiba, PR, in the years 2003 and 2012, and from Pinto Bandeira, RS, in 2012, concerning the oenological favorable aspects to the elaboration of wine. In addition, it was performed the validation of the Lorena/ELNC (80:20) medium, with regard to the detection of the killer factor, and attempts of the improvement of the technology for isolating and detecting the killer plasmids, responsible for the K+R+ phenotype in wine yeasts. The production and detection of the killer factor and the sensitive and neutral characteristics were evaluated, in triplicate, in Lorena/ELNC (80:20) solid medium. The fermentation rate and the H2S production were evaluated, in triplicate, in sulfite must medium. Strains with profile for winemaking and with killer phenotype were identified by PCR ITS1-5.8-ITS2 region amplification. The results showed that the majority of the strains presented low fermentation rate, low or absent H2S production and phenotype K-R+. In a total of 120 isolated yeasts,there were obtained , in 2012, 16 strains of Saccharomyces cerevisiae with oenological profile for the region of Pinto Bandeira. None strain, , among the 42 isolated in the Curitiba Metropolitan region in 2012, proved suitable for the elaboration of wine. Only one yeast isolated in 2003 presented suitable enologic profile. One strain of each region was chosen to vinify Pinot Noir grape to evaluate the physical and chemical characteristics of wines. There were analysed alcohol content, total reducing residual sugar, volatile acidity and pH. In five days the wines were dry, with less than 3 g.L-1 TRS, remaining only the completion of malolatic fermentation. The analysis of the volatile compounds and minerals reveled, respectively, marked concentration of some higher alcohols and high content of Zn. Thus, the autochthonous yeasts, here isolated and selected, must confer advantage over traditional spontaneous fermentation by elaborating quality wines, with the certainty that undesirable secondary metabolites are not produced. The methodology for the isolation of plasmid responsible for producing the killer factor was modified, but further studies must be made to reduce the execution time.
