dc.description.abstractResumo: As metástases são um grande problema para pacientes com câncer de mama, chegando a ser a causa de 90% dos casos de morte desta neoplasia. A proteína matriz metaloprotease 14 (MMP-14) tem uma importante contribuição no desencadeamento da migração celular nos carcinomas mamários, pela sua atividade proteolítica, e também pela ativação de outras metaloproteases solúveis capazes de degradar componentes da matriz extracelular. Sabe-se que a regulação do gene MMP14 está intimamente relacionada com aspectos epigenéticos, através da metilação de uma ilha de CpG localizada na região promotora. Estudos têm demonstrado que o padrão de metilação ao longo do corpo do gene também pode apresentar um importante papel na regulação da transcrição gênica e, para tanto, o objetivo deste estudo foi verificar o papel da metilação do corpo do gene MMP14 na regulação da expressão em diferentes linhagens tumorais de mama. As linhagens HB4aC3.6, HB4aC5.2, PMC42 e MCF-7 foram avaliadas quanto a expressão do gene estudado por RT-PCR e RT-qPCR. Duas regiões que continham ilhas de CpG, uma localizada na região promotora (-161 a +186) e outra no primeiro íntron do corpo do gene (+242 a +869), foram amplificadas e sequenciadas após o tratamento com bissulfito de sódio. Os resultados obtidos mostraram que as linhagens HB4aC3.6, HB4aC5.2 e PMC42 expressam o gene MMP14 e que suas taxas de metilação global, de ambas as ilhas de CpG avaliadas, não passam de 15%. Por outro lado, a linhagem MCF7 foi a única que não expressou o gene MMP14, e apresentou taxas de metilação global que ultrapassam 75%, em ambas as ilhas de CpG. Assim, mostrou-se uma correlação entre a ausência de expressão e alta metilação da ilha de CpG não só da região promotora, mas também da localizada no primeiro íntron. Estes resultados também permitiram a identificação de regiões diferencialmente metiladas nas linhagens de mama para padronização da técnica de MSP-PCR. A linhagem MCF7, quando submetida ao tratamento com os compostos desmetilantes (DAC) e inibidores desacetilantes de histona (TSA), passou a expressar o gene MMP14 e reduziu suas taxas de metilação global, indicando uma estreita relação entre epigenética e regulação deste gene incluindo a região intrônica. Nas regiões desmetiladas pelos tratamentos, foi possível identificar um sítio de ligação do fator de transcrição GLI3, que pode estar auxiliando na expressão de MMP14. Concluímos que a metilação da ilha de CpG localizada no íntron do gene MMP14 parece estar atuando cooperativamente com a região promotora para promover ou silenciar a transcrição do gene. Estudos de metilação no corpo do gene são recentes e, portanto, novos dados podem contribuir para entender a importância da metilação dessas regiões de DNA nos mecanismos de tumorigênese. dc.description.abstractAbstract: Metastasis is a major problem for patients with breast cancer, being the cause of 90% of deaths from this neoplasia. The protein matrix metalloprotease 14 (MMP-14) has a major contribution in the initiation of cell migration in breast carcinomas, due to its proteolytic activity, and also by the activation of other soluble metalloproteases capable of degrading components of the extracellular matrix. It is known that the regulation of the MMP14 gene is closely related to epigenetic events by methylation of a CpG island located in the promoter region. Studies have shown that the methylation pattern along the body of the gene may also have a role in regulating gene transcription and, therefore, the aim of this study was to investigate the role of methylation in the body of MMP14 in regulating the expression of this gene in different breast tumor cell lines. The expression of the gene was evaluated by RT-PCR and RT-qPCR in the HB4aC3.6, HB4aC5.2, PMC42 and MCF-7 cell lines. Two regions containing CpG islands, one located in the promoter (-161 to +186), and another region in the first intron of the gene body (+242 to +869), were amplified and sequenced after treatment with sodium bisulfite. The results showed that the HB4aC3.6, HB4aC5.2 and PMC42 lines expressed MMP14 and their rates of global methylation of both CpG islands was no more than 15%. On the other hand, the MCF7 line was the only one that did not express the MMP14 gene and presented global methylation rates exceeding 75% in both CpG islands. This showed a correlation between the absence of high expression and methylation of the CpG island not only of the promoter region but also of the first intron. These results also allowed the identification of different methylated regions in breast cancer cell lines for the standardization of MSP-PCR. The MCF7 line, when subjected to treatment with the demethylating compound (DAC) and the inhbitors of histone deacethylating compund (TSA), expressed MMP14 and reduced its rates of global methylation, indicating a close relationship between epigenetics and regulation of this gene including the intronic region. In regions demethylated by the treatment it was possible to identify a binding site of the transcription factor GLI3, which may be promoting the expression of MMP14. We conclude that methylation of the CpG island located in the intron of the MMP14 gene seems to be working cooperatively with the promoter region to permit or silence the transcription of the gene. Studies of methylation in the body of the gene are new, and, therefore, new data can contribute to understand the importance of DNA methylation of these regions in the mechanisms of tumorigenesis.
