Caracterização fitoquímica e atividades biológicas de Xylosma ciliatifolium Clos (Eichler) Flacourtiaceae (Salicaceae sensu lato)

Resumo: Na familia Flacourtiaceae (Salicaceae sensu lato), o genero Xylosma esta representado por cerca de 100 especies com distribuicao em areas tropicais e subtropicais do planeta, mas nao esta entre os mais estudados. Este trabalho teve como objetivo avaliar o potencial alelopatico, citotoxico e antibacteriano da especie Xylosma ciliatifolium, popularmente conhecida como sucara, acucara, espinho-de-judeu ou espinho-de-agulha. A partir do estudo fitoquimico das cascas das raizes fez-se o isolamento de dois compostos: manitol, um acucar poliol e de um sesquiterpeno dialdeido, sugerido como ugandensidial. Estes compostos foram relatados pela primeira vez na familia Flacourtiaceae. O oleo essencial da mesma porcao vegetal foi caracterizado por CG/EM e identificou-se 33 compostos. 3,5 dimetoxi tolueno (26,37%), limoneno (5,73%), cedranol (4,37%), E-Nerolidol (3,82%), drimenin (3,18%), a-terpineol (2,94%) e drimenol (2,74%) encontravam-se majoritarios. No ensaio alelopatico, as fracoes hexano e cloroformio das cascas das raizes e o composto XC2 (sesquiterpeno) exerceram influencia inibitoria sobre o crescimento das plantulas de Lactuca sativa. Estas mesmas amostras foram bioativas frente ao microcrustaceo Artemia salina. O teste antimicrobiano por difusao em agar revelou o potencial antibacteriano das fracoes das folhas e cascas das raizes e do isolado XC2 frente as bacterias Staphylococcus aureus e Staphylococcus epidermidis. Na avaliacao da concentracao inibitoria minima, pelo metodo da diluicao em caldo, somente as fracoes hexano e cloroformio das cascas das raizes e XC2 demonstraram atividade sobre as bacterias gram-positivas, nas concentracoes de 250, 500 e 62,5 µg/ml, respectivamente. Abstract: In the family Flacourtiaceae (Salicaceae sensu lato), the species Xylosma is represented by around 100 species distributed in tropical and subtropical areas on the planet, but it is not among the most studied ones. This paper aimed at evaluating the allelopathic, cytotoxic and anti-bacterial potential of the Xylosma ciliatifolium, commonly known as sucará, açucará, espinho-de-judeu or espinho-de-agulha. Through the phytochemistry study of the barks of the roots two compounds were isolated: mannitol, a polyol sugar and a sesquiterpene dialdehyde, suggested as ugandensidial. These compounds were firstly reported in the family Flacourtiaceae. The essential oil of the same vegetal portion was characterized as GC/MS and 33 compounds were identified. 3,5 dimetoxy tolueno (26,37%), limoneno (5,73%), cedranol (4,37%), E-Nerolidol (3,82%), drimenin (3,18%), a-terpineol (2,94%) and drimenol (2,74%) prevailed. Regarding the allelopathic test, the hexane and chloroform fractions of the barks of the roots and the compound XC2 (sesquiterpene) exerted inhibitory influence on the growth of the plantules of Lactuca sativa. These samples were bioactive regarding the brine shrimp larva Artemia salina. The antimicrobial test in agar diffusion revealed the antibacterial potential of the fractions of the leaves and barks of the roots and the isolated XC2 concerning the Staphylococcus aureus and Staphylococcus epidermidis bacteria. In the evaluation of the minimum inhibitory concentration, through broth dilution, only the hexane and chloroform fractions of the barks of the roots and XC2 showed some activity against gram-positive bacteria, in the concentrations of 250, 500 and 62.5 µg/ml, respectively.
