dc.description.abstractResumo: A dengue é, atualmente, uma das principais doenças infecciosas causadas por arbovírus, atingindo milhões de pessoas em todo mundo. Devido a fatores como: inexistência de drogas antivirais e vacinas aprovadas, sintomas iniciais que podem ser confundidos com uma série de outras infecções comuns presentes na mesma área e rápida evolução para formas graves da doença, é fundamental que se tenha disponível um teste diagnóstico rápido, inequívoco e de baixo custo, para o atendimento correto aos indivíduos infectados. Os kits diagnósticos hoje utilizados no Brasil são importados e embora apresentem resultados satisfatórios, são de alto custo. A produção de proteínas recombinantes de cepas de vírus da dengue que circulem no país e a avaliação destes insumos para o desenvolvimento de kits de diagnósticos rápidos, precisos e de custo exeqüível a realidade do país seria uma alternativa factível. Quanto à febre amarela, apesar da existência de uma vacina altamente eficaz, a prevenção e campanhas se restringem às áreas endêmicas, aumentando a vulnerabilidade da população no caso de uma epidemia. Além disso, a taxa de letalidade é muito alta e também não há nenhuma droga antiviral específica, somente tratamento de suporte. Por apresentar um modelo animal (primata) que reflete as formas clínicas observadas em indivíduos infectados, o vírus da febre amarela constitui um excelente modelo de estudo para ser estendido à outros flavivirus. Assim, o estudo funcional da protease viral do vírus da febre amarela e potenciais inibidores poderiam ajudar no desenvolvimento de drogas terapêuticas, impedindo a replicação do vírus. Baseado nestas premissas, os objetivos principais deste trabalho foram: I. Expressar as proteínas recombinantes de envelope (E), a proteína quimérica pré-membrana e envelope (prM/E) e a não-estrutural 1 (NS1), dos quatro sorotipos do DENV (DENV-1 -2, -3 e -4) utilizando e padronizando o sistema de expressão em linhagens de células S2 de Drosophila; II. Padronizar a utilização destas proteínas no desenvolvimento de kits de diagnóstico sorológico de dengue no formato ELISA, para detecção de anticorpos IgM e IgG em amostras de soro de pacientes; III. Paralelamente, produzir a protease recombinante funcional do vírus da febre amarela em sistema procariótico, assim como o controle da protease mutada no sítio ativo, para ensaios de atividade em substratos sintéticos. As proteínas prM/E, E e NS1 dos quatro sorotipos foram corretamente expressas em células S2 de Drosophila. O sistema de expressão em células S2 de Drosophila foi padronizado assim como a obtenção das linhagens de células transfectadas de forma estável com plasmídeos de expressão de diferentes proteínas recombinantes de dengue. Foram iniciados testes em formato ELISA indiretos, utilizando as proteínas recombinantes purificadas. Foi observada reatividade com soros de pacientes IgG positivos. A protease NS2B/NS3pro de febre amarela foi expressa corretamente, purificada e testada quanto a atividade catalítica. Resultados preliminares indicam que a enzima apresenta atividade em substratos sintéticos que mimetizam os sítios de clivagem da poliproteína viral nativa.pt_BR
dc.description.abstractAbstract: Currently, dengue is one of the most important mosquito-borne viral disease, afflicting millions of people in the world. Due to factors such as: the inexistence of specific antiviral drugs and approved vaccines, symptoms that can be mistaken with a series of other common infections that are present in the same area, and the rapid progression to severe forms, it is fundamental the availability of a fast, unequivocal and inexpensive diagnosis test to the proper management of patients. Although the diagnosis kits currently in use in Brazil have satisfactory results, all of them are imported and, therefore, imply an elevated cost. The production of DENV recombinant proteins based on strains of the virus present in Brazil and the evaluation of these supplies would be a feasible alternative to the development of rapid and accurate diagnosis kits, with an achievable cost to the country reality. As for the yellow fever, although there is a highly efficient vaccine, the prevention and campaigns are restrict to endemic areas, increasing the vulnerability of the population in the case of an epidemic. Moreover, the lethality rate is very high and the treatment is only supportive, due to the inexistence of specific antiviral drugs. Yellow fever virus constitutes an excellent model of study that can be extedend to other flavivirus, because it has an animal model (nonhuman primate) with clinical forms that reflects the ones observed in infected individuals. Thus, the study of the functionality of the yellow fever virus protease and its potential inhibitors could aid in the development of therapeutical drugs, hindering virus replication. Based on these premises, the main objectives of this work were: I. To express envelope (E), the chimera pre-membrane and envelope (prM/E) and non-structural 1(NS1) recombinant proteins, of the DENV four serotypes (DENV-1 -2, -3 and -4), in Drosophila S2 cells standardizing the expression of these proteins in the Drosophila expression system; II. To standardize the use of these proteins in the development of dengue serological diagnosis kits in the ELISA format, for detection of IgM and IgG antibodies in patients serum samples; III. To produce in the prokaryotic system, in parallel, the functional yellow fever virus recombinant protease, as well as its negative control, the inactive variant of protease, foruse in activity assays, with synthetic substrates. The proteins prM/E, E and NS1 of thefour types of DENV were correctly expressed in Drosophila S2cells. The Drosophila expression system was standardized and stable S2 cells that express a variety of DENV proteins were achieved. Purified recombinant proteins have been tested in indirect ELISA format and positive reaction with patient IgG serum were obtained. Yellow fever NS2B/NS3pro protease was correctly expressed, purified and its catalytic activity tested. Preliminary results indicate that the enzyme displays activity on synthetic substrates which mimic cleavage sites of the native viral poliprotein.pt_BR
