Estudo do processo biotecnológico de produção, extração e recuperação do pigmento ficocianina da Spirulina platensis

Resumo: A microalga Spirulina platensis tem sido amplamente estudada, pois apresenta altas quantidades de proteína e também biopigmentos. É considerada uma fonte potencial do pigmento-proteína ficocianina, que por sua vez, possui propriedades funcionais, como antioxidante e antinflamatória, e por ser um corante pode ser utilizado na indústria de alimentos agregando cor e valor nutricional. De forma a levantar dados importantes para a produção industrial de ficocianina, estudou-se a cinética de crescimento da microalga, como também métodos de extração otimizando a recuperação da ficocianina e purificação por precipitação, de forma a obter pureza aceitável na aplicação em alimentos. A concentração de ficocianina na microalga ao longo do tempo de cultivo, os efeitos dos diferentes métodos de rompimento celular e de pHs do solvente envolvido foram investigados, além da precipitação do pigmento em diferentes concentrações de sal para a purificação. A maior quantidade de ficocianina obtida nas culturas coincidiu com a máxima concentração celular alcançada em 15 dias de cultivo. O rompimento celular mecânico não gerou resultados satisfatórios para produção de ficocianina, comparado ao método de congelamento/descongelamento em sucessivos ciclos que resultou em concentração de ficocianina de 126 mg.g-1 biomassa seca. A purificação através da precipitação por fracionamento com diferentes concentrações de sal (NH4)2SO4] resultou em concentração de ficocianina de 30,67 mg.g-1 biomassa seca e taxa de pureza (Abs620/Abs280) igual a 2, nível satisfatório para aplicação em alimentos. Abstract: The microalga Spirulina platensis has been widely studied in the recent decades, because it presents high amounts of protein and biopigments. It is considered a potential source of the protein pigment phycocyanin, which in turn, has functional proprieties, such as antioxidant and anti-inflammatory, and as a dye it may be used in food industry adding color and nutritional value. In order to uncover useful data for the industrial production of phycocyanin, the growth of the microalga was studied, as well as methods of extraction – optimizing the recovery of phycocyanin and purification by precipitation, in order to obtain acceptable purity for use in foods. The concentration of phycocyanin in the microalga along the cultivation, the effects of different methods of cell disruption and pH of the solvent used were investigated, as well as the precipitation of pigment in different concentrations of salt for purification. The highest amount of phycocyanin obtained in the cultures coincided with the highest biomass concentration reached in 15 days of culture. The mechanical cell disruption was ineffective for phycocyanin production, as compared to freezing/thawing in successive cycles that resulted in phycocyanin concentration of 126 mg.g-1 dry biomass. The purification through precipitation by fractionation at different salt concentrations [(NH4)2SO4] resulted in a concentration of phycocyanin of 30,67 mg.g-1 dry biomass and purification rate (Abs620/Abs280) equals to 2, level adequate for use in food.
